Objective Momordicae Semen is a kind of traditional Chinese medicine commonly used in clinic.It has the effect of dispersing and detumescence,attacking poison and treating sore.The raw products of Momordicae Semen are poisonous and rich in oil.It can only be used as medicine after processing.The results showed that the content of relative active components increased,the toxicity decreased,and the antitumor effect increased.However,it is not clear about the changes of the whole components,the changes of the fatty oil components and the mechanism of the difference of the pharmacodynamics.In this study,on the basis of the previous research of the research group,the change of the whole chemical composition before and after processing was further explored.And from the perspective of fatty oil,to explore the changes of fatty oil components before and after processing,as well as the mechanism of the difference in efficacy caused by the changes.It can provide some basis for the study of the processing mechanism and anti-tumor mechanism of Momordicae Semen.Methods 1.Study on the quantitative analysis and fingerprint of the chemical components of the Momordicae Semen before and after processing The content of linoleic acid and oleic acid in fat oil before and after processing of Momordicae Semen were determined by ultra-high performance liquid chromatography(UPLC)according to the best processing technology optimized by the research group.Based on the similarity analysis,principal component analysis(PCA)and cluster analysis,the changes of components before and after processing were preliminarily determined from the perspective of quantitative and fingerprint analysis.2.Study on the differences of fatty oil chemical components in the seeds of Momordicae Semen before and after processing The optimum extraction technology of the fatty oil was optimized by orthogonal method,and the components of the fat oil before and after processing were identified by GC-MS.Based on the orthogonal partial least squares discriminant analysis(OPLS-DA)and variable importance in projection(VIP)data statistical analysis method,the differences of fatty oil components before and after processing of Momordicae Semen were determined.3.Study on the network pharmacology of the different fatty oil components in Momordicae Semen before and after processing to the liver cancer According to the different fatty oil components of Momordicae Semen before and after processing,with the help of Pub Chem,Swiss Target Prediction and BATMAN-TCM database,we can get the information related to component target.Using Gene Cards database to obtain the target information of liver cancer,the intersection of the two is the potential target of different fatty oil components in the treatment of liver cancer.With the help of STRING database,the protein interaction network of the target was obtained,and the key target was screened.Go analysis and KEGG pathway enrichment analysis of target genes were carried out by using DAVID database.The docking verification of components and key targets was carried out by Autodock vina software.Results 1.Study on the quantitative analysis and fingerprint of the chemical components of the Momordicae Semen before and after processing After processing,the relative content of linoleic acid and oleic acid increased significantly,among which linoleic acid content increased by 6.97% and oleic acid content increased by 15.85% compared with the raw product;through the similarity evaluation software of traditional Chinese medicine chromatographic fingerprint,22 fingerprint peaks of 20 batches of samples before and after processing were determined.The results of cluster analysis,PCA analysis and OPLS-DA analysis showed that there were some differences in the similarity of fingerprints of the samples before and after processing of Momordicae Semen.The results showed that the chemical components of the Momordicae Semen changed in quality and quantity before and after processing.2.Study on the differences of fatty oil chemical components in the seeds of Momordicae Semen before and after processing Orthogonal design was used to optimize the extraction process of fatty oil.The best extraction process is that the ratio of material to liquid is 1:40,the extraction time is 6h,and the extraction temperature is 85 ?.According to this technology,the fatty oil of Momordicae Semen was extracted before and after processing,and its components were detected by GC-MS.14 kinds of fatty oil components were identified,11 kinds of fatty oil components increased and 3 kinds of fatty oil components decreased after processing.The results of OPLS-DA showed that there were differences in the fatty oil components of Momordicae Semen before and after processing.Through VIP analysis,a total of 8 different fatty oil components of Momordicae Semen before and after processing were selected,including azelaic acid,myristic acid,palmitic acid,oleic acid,stearic acid,linoleic acid,10,12-octadecadienoic acid and eleostearic acid.3.Study on the network pharmacology of the different fatty oil components in Momordicae Semen before and after processing to the liver cancer According to the results of 8 kinds of different fatty oil components before and after processing,548 related targets of 8 kinds of different fatty oil components before and after processing were obtained by using the relevant database,and after eliminating the repeated targets,172 targets were obtained.Through the intersection of 1658 related targets of liver cancer,82 targets of different fatty oil components of Momordicae Semen were obtained.Through protein-protein interaction,14 key targets were screened out.Through molecular docking,it has been proved that linoleic acid and oleic acid can well bind to mapk3,MAPK14,PPARG and other targets,which are mostly concentrated in PPAR signaling pathway,TNF signaling pathway,NOD-like signaling pathway and Toll-like receptor signaling pathway.From the point of view of fatty oil components,it is suggested that the mechanism of the difference of pharmacodynamic effect before and after the processing may be related to the effect of linoleic acid and oleic acid on related targets and their corresponding pathways,thus affecting the occurrence and development of diseases.Conclusion Before and after processing,the content of linoleic acid and oleic acid increased,and there were some differences in fingerprints,which initially proved that there were differences in quality and quantity of the whole components before and after processing of Momordicae Semen.In addition,the changes of fatty oil components before and after the processing of Momordicae Semen were obvious.Combined with the results of network pharmacology,it was preliminarily shown that the different fatty oil components before and after the processing of Momordicae Semen can affect the occurrence and development of the disease by acting on the related targets and corresponding pathways of liver cancer,thus resulting in the difference in efficacy.From the point of view of fatty oil components,it provides a certain basis for the study of the processing mechanism and anti-tumor mechanism of Momordicae Semen.
