Gymnemic acids is a class of triterpenoid saponins isolated from Gymnema Sylvestre.It has many biological activities,such as inhibiting oleic acid absorption in small intestine,inhibiting triphosphate dehydrogenase activity,inhibiting alpha-glucosidase activity,inhibiting sodium-dependent glucose transporter-1 activity,inhibiting cancer and so on.It is widely used to treat diseases caused by metabolic disorders.It is hypoglycemic and lipid-lowering in Athyllum fortunei.Main active ingredients.At present,there are more and more studies on the activity of pachytenic acid,but the main focus is on the hypoglycemic activity of pachytenic acid.The mechanism of pachytenic acid regulating blood lipid is still unclear.This paper mainly evaluates the regulating effect of atropic acid on blood lipid level through animal experiments,and preliminarily studies its mechanism of regulating blood lipid.Male Syrian golden hamsters were fed with high-fat diet to establish a high-fat model.They were fed with high-fat diet for one week before the experiment.Then they were randomly divided into five groups:normal control group,model control group and low,medium and high doses of Gymnemic acid group.Normal control group was fed with normal diet.The other groups were fed with high-fat diet for 8 weeks.The body weight was weighed once a week and blood was taken from the orbital vein every two weeks to detect the level of blood lipid in the serum of hamsters at different stages.Eight weeks later,femoral artery blood was taken and killed.Antioxidant enzyme activity,MDA content,AST level,ALT level,body fat and organ index in serum of hamsters were measured,and liver tissue morphology was observed.Result:Gymnemic acid could effectively inhibit the increase of total cholesterol(TC),triglyceride(TG)and non-high density lipoprotein cholesterol(non-HDL-C)in the serum of high-fat-fed hamsters,and the inhibition effect of TG was the best.Gymnemic acids could effectively inhibit the increase of liver organ index,serum ALT and AST,and reduce the risk of liver steatosis.It can also increase the activity of antioxidant enzymes in serum and reduce the accumulation of MDA in serum.On the basis of the above physical and chemical experiments,the mechanism of regulating blood lipids was preliminarily studied by enzyme-linked immunosorbent assay(ELISA)and polymerase chain reaction(PCR).Enzyme-linked immunosorbent assay(ELISA)was used to detect the levels of HMGCR,LDLR,AC AT and CYP7A1,the key enzymes involved in the regulation of cholesterol metabolism in hamster serum,and the expression levels of HMGCR,ACAT,CYP7A1 and other related genes in hamster serum were detected by PCR.Enzyme-linked immunosorbent assay(ELISA)results showed that,on the one hand,it could reduce cholesterol synthesis in high-fat hamsters by inhibiting the activity of HMGCR,on the other hand,it could increase cholesterol excretion by increasing the activity of CYP7A1,so as to effectively regulate the blood lipid level of hamsters.Polymerase chain reaction(PCR)results showed that Gymnemic acids could inhibit the increase of blood lipid level in high-fat diet hamsters by inhibiting the synthesis of cholesterol,reducing the absorption of cholesterol in small intestine and accelerating the excretion of cholesterol,and increasing the content of LDLR in liver of hamsters,so as to inhibit the increase of blood lipid level in high-fat diet hamsters.
