Objective:By detecting the apoptosis rate and the expression of apoptotic factors Caspase-3,Fas,and P53 in lumbar degenerative hypertrophic ligamentum flavum tissue,we can lay the foundation for understanding the signaling pathways involved in lumbar ligamentum flavum cell apoptosis.Methods: The experimental group was taken from 50 patients with lumbar spinal stenosis who underwent surgical decompression.All the ligamentum flavum were confirmed to be degenerative hypertrophic ligamentum flavum by MRI measurement before and after measurement,including 22 males and 28 females,age 32-74 years old,average 54.46 years old.The control group was obtained from 30 patients with lumbar disc herniation and burst lumbar fracture who underwent surgical decompression.All the ligamentum flavum were confirmed to be non-hypertrophic ligamentum tissue by MRI measurement before and after measurement,including 19 males and 11 females,age 19-67 years old,average 47.27 years old.The material was taken from the L2-S1 segmental ligamentum flavum.The thickness of the ligamentum flavum of the two groups was measured,the apoptotic cell rate in the ligamentum flavum was detected by TUNEL staining,and the expression levels of the apoptotic factors Caspase-3,Fas and P53 were detected by SP immunohistochemical staining.Results:(1)The thickness of the experimental group is 4.5-8.0 mm,with an average of 5.63 mm,and the thickness of the control group is 2.3-4.0 mm,with an average of 3.56 mm.The difference between the two groups is statistically significant;(2)TUNEL staining showed that the average apoptotic rate of the experimental group was(37.80±3.04)%,which was significantly higher than that of the control group [(13.18±1.34)%;t=41.83,P < 0.001];(3)The immunohistochemical staining of SP showed that the positive expression percentages of Caspase-3,Fas and P53 in the ligamentum flavum were all 10% in the experimental group,while the positive percentages were 13.3%,16.7%,and 10% in the control grou,respectively.There was a significant difference between the two groups(P < 0.001).Conclusion: Apoptosis increased in degenerative and hypertrophic ligamentum flavum tissue of lumbar spine.The expression levels of apoptotic factors Caspase-3,Fas and P53 in degenerative and hypertrophic ligamentum flavum were up-regulated compared with normal ligamentum flavum.
