Objective: Detection of hypoxia-inducible factor-1 ?(HIF-1?)and its target gene vascular endothelial growth factor(VEGF)and B lymphoma / leukemia-2(bcl-2)/adenovirus E1 B 19000 interacting protein 3(BNIP3)The expression of IH in infants and young children and the expression of HIF-1? in the serum of IH children before and after propranolol treatment were investigated to explore the role of HIF-1?-mediated hypoxia-related pathway in IH.Methods: IH paraffin tissue was surgically removed from the Department of Pathology,Xinjiang Uygur Autonomous Region People's Hospital from January 2015 to January 2018.Thirty cases were in the proliferative phase,30 cases were in the regression phase,and 10 were normal skin tissue.The expression of HIF-1?,BNIP3,and VEGF in the proliferative,regressive,and normal tissues were detected by immunohistochemical methods.Enzyme-linked immunosorbent assay(ELISA)was used to detect before and after propranolol treatment.The changes of serum HIF-1?concentration before and after treatment were observed,and its role in the occurrence and development of IH was analyzed.Results: The average optical density values of HIF-1?,VEGF and BNIP3 in IH paraffin tissue during proliferation period were 0.3733±0.0978,0.275±0.037 and 0.232±0.061 respectively,and the average optical density values in paraffin tissue during regression period were 0.3207±0.0597,0.226±0.04 and 0.186±0.059,the average optical density values in paraffin tissue of normal control group were0.267 ± 0.069,0.226 ± 0.04 and 0.158 ± 0.058,respectively,and the average optical density values of paraffin tissue in proliferation period were significantly higher than The regression period(P<0.05),the regression period was not statistically different from normal tissues(P>0.05).The concentrations of HIF-1? in the serum of children before and after 12 weeks of propranolol treatment and the normal control group were 0.228±0.069mg/L,0.174 ± 0.041mg/L and 0.148 ± 0.021mg/L,the results showed The concentration of HIF-1? before propranolol treatment was significantly higher than that at 12 weeks after treatment,the difference was statistically significant(P<0.05),and there was no significant difference at 12 weeks after treatment compared with the normalcontrol group(P> 0.05).Conclusion: HIF-1? and its target genes BNIP3 and VEGF play an important role in the occurrence,development and regression of IH.Propranolol inhibits the growth of hemangioendothelial cells by inhibiting the expression of HIF-1?,and promotes the regression of IH.To achieve the purpose of treating IH.
