Objective: To explore the clinicopathological significance of CXCL6 expression in patients with esophageal cancer,and the effects of CXCL6 on epithelial-mesenchymal transition(EMT)and epithelial-mesenchymal transition.To clarify the molecular mechanism of CXCL6 involved in the regulation of malignant phenotype in esophageal cancer cells.Methods: 1)Clinical phenotype: the expression of CXCL6 in 105 cases of esophageal cancer was detected by Immuno histo chemical(IHC).The correlation between the expression of CXCL6 and the prognosis and clinicopathological significance of the patients was analyzed.2)Cell function test and mechanism: the effects of CXCL6 on migration,invasion,proliferation and Apoptosis of esophageal cancer cells were detected by MTT Assay,Wound-healing Assay,Transwell Assay and Apoptosis Assay.The effect of CXCL6 on the expression of EMT-related marker protein in esophageal cancer cells was detected by Western blot.To detect the effect of CXCL6 on PD-L1 and p-STAT3 and to explore the relationship between PD-L1 and STAT3 pathways.Preliminarily investigate the molecular mechanism of CXCL6 involved in the regulation of malignant phenotype of esophageal cancer cells.3)Animal experiments: the esophageal cancer cells were transfected with lentivirus,and the overexpression and knockdown cell lines of esophageal cancer were constructed respectively.Subcutaneous tumorigenesis model was established by subcutaneous injection to detect tumor growth in 40 nude mice,and metastasis model was established by tail vein injection to detect metastasis in 40 nude mice.Results: 1)Clinical phenotype: immunohistochemical results showed that the expression of CXCL6 in esophageal cancer tissues was significantly higher than that in adjacent normal tissues,and the difference was statistically significant(P<0.05).The results of clinicopathological analysis showed that CXCL6 expression was statistically significant with the differentiation degree of esophageal cancer patients(P<0.05).In addition,Kaplan-Meier(K-M)survival curve analysis showed that the positive co-expression of CXCL6 and PD-L1 was statistically significant with the overall prognosis of patients(P<0.05).2)Cell function test and mechanism: Western blot results showed that CXCL6 promoted the expression of PD-L1 by activating the STAT3 pathway,thereby promoting migration,invasion,proliferation,EMT and inhibiting apoptosis of esophageal cancer cells.3)Animal model: experimental results in animals showed that CXCL6 promoted tumor growth and lung metastasis.Conclusion: CXCL6 promotes the expression of PD-L1 to regulate the malignant phenotype of esophageal cancer cells by activating the STAT3 signaling pathway.CXCL6 is expected to become a new target for the treatment of esophageal cancer and another indicator for the evaluation of clinical tumor prognosis.
