Objective: To study the role of NLRP3 inflammasome in the apoptosis of bladder mucosa cells in diabetic rats,and to discuss the relationship between NLRP3 inflammasome and diabetes cystopathy.By studying the role and changes of NLRP3 inflammasome in the development of diabetes cystopathyr,we can provide a new basis and target for the etiology research and treatment of diabetes cystopathy.Methods: 25 healthy female Wistar rats were randomly divided into two groups: diabetic group and control group.The diabetic group was induced by intraperitoneal injection of streptozotocin.Blood samples were collected 72 hours after injection,and the random blood glucose >16.7 mmol / l as the indicator of the models was successfully made.The control group was intraperitoneally injected with pH4.5,0.1mol/l citric acid sodium citrate buffer at the same dose.After the model was successfully made,the blood glucose was measured and weighed regularly.The bladder tissue structure was observed under the microscope for 16 weeks.TUNEL technique was used to detect the apoptosis level of bladder mucosa The levels of NLRP3,pro caspase 1 and cle caspase 1 were detected by Western blot,and IL-1 ? was detected by ELISA technique.Results: In the control group,the morphology of mucous membranes were normal without hyperplasia,the fibrous connective tissue of hypodermic lamina was arranged regularly,and the muscular layer were arranged orderly and closely.A small number of apoptotic cells were observed.In the diabetic group,there were obvious hyperplasia of the epithelium of the mucosa layer,hyperemia and edema of the hypodermic lamina propria,infiltration of a large number of acute and chronic inflammatory cells in the mucosa epithelium and the lamina propria;the smooth muscle bundle of the muscle layer was loose and disordered,some of the muscle bundles were broken,and the space was widened,and a large number of small blood vessels were congested and a small number of chronic inflammatory cells wereinfiltrated.A large number of apoptotic cells were observed.Conclusion: The levels of NLRP3,pro-caspase 1,cle-caspase 1 and IL-1? increased significantly(P < 0.05).The activation of NLRP3 inflammasome is involved in the apoptosis of bladder mucosa cells and is related to the formation of diabetes cystopathy.
