Therapeutic Effect Of Osteoking On Tree Shrew With Osteoporotic Fracture

Objective:To investigate the effects of Osteoking(OK)on body weight,body composition,blood physiological and biochemical indexes and organ coefficient of tree shrews in postmenopausal osteoporotic fracture model.Methods:Totally 30 female western Yunnan female tree shrews were randomly divided into the control group,the model group and the treatment group,with 10 in each group.The same volume of omentum as the uterus and ovary was excised in the control group.The ovaries and uterus were removed in model group and treatment group,and osteoporosis was confirmed 6 months later,then osteoporotic fracture(OPF)model was established by making tibial fracture of right hind limb.From the next day of fracture,the treatment group was given OK orally once every two days,while the model group and the control group were given the same amount of saline.The body weight,body Composition,blood physiological and biochemical indicators and organ coefficients were detected after treated for 3 months.Results:The body weight of the model group was significantly higher than that of the treatment group and the control group(P<0.01),but there was no significant difference between the treatment group and the control group(P>0.05).Compared with the control group,the BMD of the whole body and various parts of the OPF model group was significantly reduced;the whole body(including the head and no head),the spine and lumbar vertebrae,the mineral content was significantly reduced,and the percentage of adipose tissue was significantly increased;the pelvic bone mineral content was significantly reduced,the percentage of muscle tissue was significantly decreased,and the percentage of adipose tissue was significantly increased.The percentage of adipose tissue in the lumbosacral,left pelvis and right pelvis was significantly increased,and the difference was statistically significant(P<0.05 or P<0.01).Compared with the model group,the bone density of the spine,lumbar vertebrae,lumbosacral humerus,pelvis and left pelvis in the treatment group increased significantly;the percentage of the whole body(including the head and no head),the spine and the pelvis muscle tissue increased significantly.The percentage of adipose tissue was significantly decreased.The percentage of adipose tissue in lumbar vertebrae,lumbosacral humerus,left pelvis and right pelvis was significantly lower(P<0.05 or P<0.01).When OK was used to treat OPF tree shrew,the BMD improvement rate of lumbar vertebrae was higher than that of other parts.Compared with the control group,the blood physiological indexes(percentage of monocytes,absolute value of monocytes,white blood cell count,percentage of lymphocytes,absolute value of lymphocytes)and some blood biochemical indicators(cholesterol,glucose,uric acid,high density lipoprotein,low density lipoprotein,apolipoprotein B)of tree shrew in the model group decreased significantly;however,the other blood biochemical indicators(alanine aminotransferase,creatine kinase isoenzyme,creatinine,magnesium ion,aspartate aminotransferase,serum creatine phosphokinase,total iron binding capacity,urea nitrogen,phosphorus)were significantly increased(P<0.01).Compared with the model group,the blood physiological indexes(percentage of monocytes,absolute value of monocytes,percentage of lymphocytes,absolute value of lymphocytes,white blood cell count)and blood biochemical indicators(glucose)were significantly increased in the treatment group(P<0.01 or P<0.05);while some blood biochemical indicators(alanine aminotransferase,creatine kinase isoenzyme,creatinine,magnesium ion)were significantly decreased,and the difference was statistically significant(P<0.01 or P<0.05).Compared with the control group,the spleen coefficient of the model group was significantly lower than that of the control group(P<0.01);compared with the model group;the spleen coefficient of the treatment group was significantly higher than that of the model group(P<0.05).Conclusions:OK can regulate the lipid metabolism disorder,reduce the percentage of body fat,increase the percentage of muscle of OPF tree shrew,and inhibit obesity caused by OPF.OK can also improve BMD in all parts of the body of OPF tree shrew,especially for the lumbar spine.In addition,OK can also improve the immunity and the functions of heart,liver and kidney of OPF tree shrew.
