BackroundBecause of the development and progress of modern society and the rising level of medical care,the elderly people in the total population is getting larger and larger,and the social and medical problems it brings are becoming more and more obvious.The contradiction of aging society is becoming increasingly prominent.One of the more important diseases,the influences of osteoporosis can't be ignored,and it has gradually attracted widespread attention from the government,scientific research institutions and medical workers.As a serious complication of osteoporosis,the proportion of social public medical services expenditure caused by fractures has also gradually expanded.According to reports,85% of the cost of fracture treatment is closely related to osteoporosis,and the cost of treatment it generates is more than 100 billion.The huge increase of the scale of the elderly people,accompanied by the advancement of medical and scientific and technological levels,requires greater progress and development.Among them,SIRT1 and PGC-1?,which are related to aging of the population,have been confirmed to be closely related to bone metabolism.ObjectiveThe main research purpose of this article is to further study the expression of the related aging genes SIRT1 and PGC-1? in the osteoporosis population in order to understand the detailed mechanism and relationship.According to the research results and data analysis,more detailed understanding of the related genes on the skeletal system The influence of metabolism provides directions for the early diagnosis,prevention and treatment of osteoporosis.MethodsSixty-four elderly patients with osteoporosis admitted to our department from October 2018 to December 2019 were selected for DXA examination during hospitalization.They were seprated from two groups: normal bone mass group and osteoporosis group by osteoporosis diagnostic criteria.Blood samples were pretreated separately,and then real-time fluorescence quantitative PCR(q RT-PCR)was processed.After statistics and results analysis,the differences and problems between the two groups of patients were compared.ResultThe results of q RT-PCR test showed that compared with the samples with normal bone mass,the expression of SIRT1 osteoporosis group was significantly reduced(P <0.01).The upstream and downstream mediated indicators affected by SIRT1 have increased or decreased to varying degrees,and the statistical results also showed correlation.In the serum samples of osteoporosis group,the content of PGC-1? also decreased significantly compared with the group with normal bone mass(P <0.01).Conclution1.The expression of SIRT1 in serum samples of osteoporosis group was significantly lower than that of normal group;2.The expression level of PGC-1? in serum samples of patients with osteoporosis group was lower than that of normal group;3.The substance content of bone formation and bone differentiation affected by SIRT1 has increased and decreased to varying degrees;4.The influence of both SIRT1 and PGC-1? on bone metabolism is obviously related.By further deepening the research on the two,we can provide reference suggestions for the pathogenesis and preventive treatment of osteoporosis.
