Cancer is one of the most prominent diseases that threaten human health in the world.The large number of different forms of cancer has limited people's research on the pathogenesis and treatment of cancer.Pan-cancer analysis aims to study the similarities and differences between different cancer types.By combining data from multiple cancers,we can better understand the origin of cancer cells and the process of carcinogenesis,thereby finding a universal treatment method.Metabolism reprogram-ming is the immediate cause of tumor transformation and development,so analyzing cancer from a metabolic perspective is of great significance for cancer diagnosis and treatment.Metabolic network reconstruction can characterize the metabolic state of diseased cells.Pan-cancer analysis based on the metabolic network reconstruction method is beneficial to systematically understand the internal working mechanism of cancer and obtain more robust analysis results.These results can provide guidance for biological experiments and drug discovery.In this dissertation,the metabolic network reconstruction method is used to analyze the metabolic characteristics of tumors originating in different tissues from multiple per-spectives.The main works include:(1)We give a pan-cancer analysis method based on cancer work network.The Cancer Work Network is based on the highly expressed core response set of cancer and can accurately represent the metabolic state of tumor cells.We successfully obtained 27 cancer delivery reactions and 7 potential drug targets.The results help to understand the common characteristics of cancer metabolism,and also provide guidance for biological experiments.(2)A pan-cancer analysis method based on the differential metabolic network is proposed.The differential metabolic network is built on the basis of differentially expressed genes,which effectively eliminate the in-fluence of the interference of tissue metabolic characteristics.The experiment obtained differentially expressed genes,reactions and metabolic pathways of 17 types of cancer.The analysis results help to understand the mechanism of cancer and promote the re-search of cancer treatment methods.(3)We give a pan-cancer analysis method based on gene chip data.Aiming at the shortage of available samples in the current pan-cancer metabolism research,this paper designs a complete set of microarray data acquisition and preprocessing processes based on the GEO database,and finally obtains more than 20,000 microarray data samples that can be used directly.Based on these data,a pan-cancer metabolic analysis was conducted and new cancer knowledge was discovered.We obtained cancer-related metabolic pathways,potential pan-cancer drug targets and tumor suppressor genes of 18 cancers.The experimental results proved the effective-ness of the method and high-quality microarray data sets can also provide support for other related research.
