Objective Observe the expression of NLRP3 inflammasome in lung cancer and its relationship with clinicopathological characteristics,and explore its potential role in the development of lung cancer.Methods One hundred and twenty paraffin-embedded specimens were collected from patients undergoing surgical treatment or puncture in our hospital from March 2014 to January 2018 and confirmed by postoperative pathology as lung cancer,including 45 cases of squamous cell carcinoma(SCC)and 55 cases Adenocarcinoma(ADC)and 20 cases of small cell lung cancer(SCLC)(of which 60 cases have tissue adjacent to the cancer).At the same time,the personal information and detailed clinical data were collected,andthe componentsof NLRP3 inflammasomewhich including NLRP3,ASC,caspase-1,and its downstream active factorsIL-1?and IL-18 were detected by immunohistochemical staining(IHC)method in tissues and their relationship with clinical features of lung cancer were detected,and the correlation of protein expression was explored.Results 1.The positive expression rates of the componentsof NLRP3 inflamm--asomewhich including NLRP3,ASC,caspase-1,and its downstream active factors IL-1? and IL-18 in lung cancer tissues were 65.0%,55.0%,65.8%,59.2% and 55.8%,respectively,which were significantly higher than thosein adjacent tissues(P<0.05).2.The positive expressions of NLRP3 in ADC,SCLC and SCC were 76.4%,65.0% and 51.1%,There was a statistical difference between the groups(P<0.05).The expression rates of NLRP3 in the groups with lymph node metastasis,di stant metastasis and low differentiation were 83.3%,95.5% and 76.8%,respectivel y,which were higher than those in the groups without lymph node metastasis(42.6%),distant metastasis(58.2%)and high and Moderately differentiation(54.7%),s howing statistical differences(P<0.05).In addition,among the 42 patients with stag e III+IV,NLRP3 was positively expressed in 35 patients(83.3%),showing statistical differences(P<0.05);The positive expressions of ASC in ADC,SCLC and S CC were 72.5%,70.0% and 38.5%,There was a statistical difference between the groups(P<0.05).The expression rates of ASC in the group with lymph node met astasis and the group with low differentiation were 66.7% and 67.9%,respectively,which were higher than those in the group without lymph node metastasis(40.7%)and the group with high and Moderately differentiation(43.8%),showing stat istical differences(P<0.05).In addition,of the 42 patients with stage III+IV,30(71.4%)had positive ASC expression,showing a statistical difference(P<0.05),ther e was no correlation between ASC and distance transfer(P>0.05);The positive e xpressions of caspase-1 in ADC,SCLC and SCC were 80.0%,55.0%,and 53.3%,respectively,which were statistically different(P<0.05);the expression rate of cas pase-1 in lymph node metastasis and poorly differentiated groups were 80.3% and80.4%,respectively,which were higher than those without lymph node metastasis(48.1%)and high and Moderately differentiated group(53.1%),and there was a statistical difference(P <0.05).caspase-1 was not related to TNM stage and dista nt metastasis(P> 0.05);The positive expression of IL-1? in stage II+IV(78.6%)was significantly higher than that in stage ?+?(44.8%),which was statistically di fferent(P<0.05);the expression rate of IL-1? in the lymph node metastasis and p oorly differentiated group were 72.7% and 69.6%,which were higher than those without lymph node metastasis(42.6%)and high and Moderately differentiation(50.0%),and there was a statistical difference(P<0.05);IL-1? was not related to th e pathological type and distant metastasis(P>0.05);the positive expressions of IL-18 in ADC,SCLC,and SCC were 65.5%,65.0%,and 40.0%,respectively,with s tatistical differences(P<0.05);in 66 patients with lymph nodes Among metastatic patients,43 patients had positive IL-18 expression(65.2%);among 56 poorly diff erentiated patients,40 patients had positive IL-18 expression(71.4%),all of whic h were statistically different(P<0.05).there was no correlation between IL-18 and TNM stage and distant metastasis(P>0.05).The expressions of the abovefive prot eins had no significant correlation with age,gender,and tumor size(P>0.05).3.F urther analysis of the relationship between the three components of NLRP3 inflammasome and the expression of IL-1? and IL-18 in lung cancer tissues showed tha t there was a positive correlation between the above protein expression levels(P<0.05).Conclusion 1.Levels of NLRP3,ASC,caspase-1,IL-1?,and IL-18 are up-reg ulated in lung cancer tissues.2.The expression levels of NLRP3,ASC,caspase-1,IL-1?and IL-18 were related to the pathological type,TNM stage,differentiation a nd metastasis of lung cancer.3.There was a positive correlation between theexpres sions of the three components of NLRP3 inflammasome and the expression of IL-1? and IL-18.
