Objective:It was found that LD ointment Tuina has the effect of Promoting the growth and development of children with cerebral Palsy(CP),but the molecular mechanism is unclear.In this experiment,the expression of somatostatin(SS)and growth hormone releasing hormone(GHRH)Protein in Peripheral and central of cerebral Palsy model was detected,To exPlore the molecular mechanism of LD ointment Tuina to Promote the growth and development of CP rats,and lay the experimental foundation for the clinical Promotion of invention Patent of LD ointment Tuina.Method:According to the research standard,60 male rats were selected from 115 normal SD rats.They were randomly divided into the model group(n=40),the blank control group(n=10)and the sham oPeration group(n=10).The model group used the left common carotid artery ligation combined with hyPoxia on the third day of birth(P3)to PrePare the cerebral Palsy PuP model.The sham oPeration control group only cut the neck skin of the rats and seParated the left neck 30 SD rats were randomly divided into three sub groups:LD ointment Tuina group(n=10),skin oil group(n=10),model control group(n=10).LD ointment Tuina was used as the massage medium in the group of LD ointment Tuina and the control group was used as the massage medium.The blank control group,the model control group and the sham oPeration control group were not treated by massage,but were kePt in seParate cages with the female rats in the LD ointment Tuina group and the moisturizing oil massage group.The body weight,body length+tail length,eye oPening time time of the young rats were observed and recorded at the same time Point in each group.Gait analysis and negative earth moving test were carried out.The Protein exPression of SS and GHRH in PeriPheral blood,central hyPothalamus were measured.One way ANOVA and indePendent samPles were used for SPSS statistical software P<0.05,the difference was statistically significant;when P<0.01,the difference was statistically significant.Conclusion:1.After the establishment of the model,the righting exPeriment was carried out to verify the establishment of the model.The righting time of the model group was 12.91±6.25s,comPared with the sham oPeration control group(1.42±0.34s),P<0.01,the difference was statistically significant.ComPared with the blank control grou(1.64±0.29s),the difference was statistically significant(P<0.01).2.After 4 weeks of massage,the weight of LD ointment Tuina group was 114.92±2.78g,comPared with that of the control grou(99.93±4.42g),P<0.01,the difference was statistically significant.ComPared with the model control grou(87.79±3.26g),the difference was statistically significant(P<0.01).The difference was statistically significant(P<0.05).The difference was statistically significant(P<0.05).After 4 weeks of massage,the body length+tail length of LD ointment Tuina group was 30.93±0.34cm,comPared with the control group(28.80±0.54cm),P<0.01,the difference was statistically significant.ComPared with the model control group(26.54±0.80cm),the difference was statistically significant(P<0.01).There was no significant difference between LD ointment Tuina group and sham oPeration control group(30.57±0.68cm),P>0.05.There was no significant difference between LD ointment Tuina group and blank control group(30.54±0.72cm),P>0.05.The oPening time of LD ointment Tuina group was 14.30±0.48d,comPared with the control group(14.90±0.31d),P<0.05,the difference was statistically significant.ComPared with the model control group(16.30±0.48d),the difference was statistically significant(P<0.05).The difference was statistically significant(P<0.01).The difference was statistically significant(P<0.01).3.After 4 weeks of massage,the motor function of young rats was evaluated by gait analysis.The ground moving sPeed of LD ointment Tuina group was(5.02±0.80s),comPared with that of the control group(3.77±0.56s),P<0.01,the difference was statistically significant.The difference between LD ointment Tuina group and the model control group was(3.60±0.51s),P<0.01,the difference was statistically significant.There was no significant difference between LD ointment Tuina group and sham oPeration control group(4.58±0.73s),P>0.05.There was no significant difference between LD ointment Tuina group and blank control group(5.15±0.57s),P>0.05.ComPared with the control group(9.67±0.33mm),the difference was statistically significant(P<0.01).ComPared with the model control group(8.11±0.54mm),the difference was statistically significant(P<0.01).There was no significant difference between LD ointment Tuina and sham oPeration(12.11±1.01mm),P>0.05.There was no significant difference between LD ointment Tuina group and blank control group(11.41±1.17mm),P>0.05.After 4 weeks of massage,the motor resPonse ability of young rats was detected by negative trend.The difference between LD ointment Tuina group(1.35±0.27s)and skin oil control group(1.96s±0.18s)was statistically significant(P<0.05).The difference between LD ointment Tuina group and model control group(2.37±0.17s)was statistically significant(P<0.01).There was no significant difference between LD ointment Tuina group and sham oPeration control group(1.43±0.16s),P>0.05.There was no significant difference between LD ointment Tuina group and blank control group(1.35±0.12s),P>0.05.4.After four weeks of massage intervention,the exPression of SS in LD ointment Tuina group was 91.248±24.457ng/ml,comPared with the control group(110.664±7.814ng/ml),P>0.05,there was no significant difference.ComPared with the model control group(122.520±59.617ng/ml),the difference was statistically significant(P<0.05).There was no significant difference between LD ointment Tuina group and sham oPeration control group(90.188±28.466ng/ml),P>0.05.There was no significant difference between LD ointment Tuina group and blank control group(88.314±22.27ng/ml),P>0.05.ComPared with the control group(17.107±1.311ng/ml),the exPression of GHRH was 17.804±0.794ng/ml(P>0.05).ComPared with the model control group(10.829±0.404ng/ml),the difference was statistically significant(P<0.01).There was no significant difference between LD ointment Tuina and sham oPeration control group(18.328±0.897ng/ml),P>0.05.There was no significant difference between LD ointment Tuina group and blank control group(18.453±1.281 ng/ml),P>0.05The ratio of central SS expression and internal reference Protein in LD ointment Tuina group was 0.0245±0.008,comPared with that in skin oil control group(0.716 ±0.041),P>0.05,there was no significant difference.ComPared with the model control group(0.967±0.127),the difference was statistically significant(P<0.01).The difference was statistically significantP<0.01.There was significant difference between the control group(0.390±0.008)and the control group(P<0.01)The ratio of GHRH and internal reference Protein was 1.005±0.101 in the group of LD ointment Tuina and moxibustion,P>0.05 comPared with the control group(0.850±0.082).ComPared with the model control group(0.420±0.070),the difference was statistically significant(P<0.01).Conclusion:1.LD ointment Tuina can Promote the growth and development of CP rats.The experimental results are consistent with the Previous rePorts2.LD ointment Tuina can imProve the motor ability of CP Rats,the experimental results are consistent with the Previous literature.3.The effect of LD ointment Tuina on Promoting the growth and develoPment of CP young rats and imProving the motor function may be realized by reducing the SS Protein in PeriPheral center and Promoting the exPression of GHRH Protein in PeriPheral and central,and then regulating the release of growth hormone(GH),which Provides exPerimental basis for the treatment of CP with LD ointment Tuina.
