| Low immunity is a sub-health state that is common in modern society.In this state,the human body is susceptible to invasion by pathogenic bacteria.The immune response,an important protective barrier,has a direct relationship with the health of the human body,and the strength of the immune system is very important for human health.Functional substances screening out from natural foods with immunomodulatory peptides can be made into health products or nutritional products to improve human immunity.Chicken embryo eggs,as a folk medicinal dietary supplement,have a long history of being eaten due to their rich nutrition and smooth and tender taste.Research reports have shown that chicken embryo egg homogenate,chicken embryo egg water solubles,and chicken embryo egg fat solubles all can promote the immunity.In this paper,chicken embryo eggs are used as raw materials to obtain chicken embryo peptides of different molecular weights through protein extraction,optimized hydrolysis process,and chicken embryo peptide separation,and to explore its role in enhancing and regulating immunity.In this paper,13-day-old chicken embryo eggs are used as raw materials to obtain crude chicken embryo protein.The enzymatic hydrolysis effects of five proteases are explored(subtilisin,alcalase,papain,bromelain,and trypsin)through single factor and response surface experiments.After optimizing the process of bromelain hydrolysis of chicken embryos,it was found that the optimal hydrolysis conditions were: pH6,enzyme to substrate ratio 3%,temperature 45℃,bromelain,time 7 h.On the basis of optimizing the enzymolysis process of chicken embryo eggs,chicken embryo peptides were obtained under the optimal process conditions.Using ultrafiltration and dialysis techniques,four groups of chicken embryo polypeptides with molecular weights less than 1 kDa,1-5kDa,5-10 kDa,and greater than 10 kDa were obtained.Adjust the concentration of the four components,give the mouse macrophages,and the blank control group give the complete medium.Compared with the control group,the four groups of chicken embryo peptides of different concentrations all promote the proliferation of mouse macrophages and enhance the phagocytic activity of macrophages.The mouse macrophages RAW264.7 were divided into three groups,which were given complete medium,lipopolysaccharide(LPS),lipopolysaccharide and chicken embryo peptides of different concentrations.LPS can inhibit cellular immune function,promote the release of macrophage NO,and promote the gene expression of related immune factors(iNOS,IL-6,IL-10,TNF-α,TLR4).Compared with the lipopolysaccharide stimulation group,the chicken embryo peptide stimulation group showed a decrease in macrophage NO release,and the gene expression of iNOS mRNA,IL-6 mRNA,IL-10 mRNA,TNF-α mRNA,and TLR4 mRNA callback,indicating chicken embryo peptides can promote immunity. |
