Objective To observe the effect of Qingre Huoxue Fang on the general morphological and pathological changes of rheumatoid arthritis rats,and to detect the effects of NLRP3 inflammatory bodies and the expression of related factors IL-1?and IL-18,and to explore Qingre Huoxue Recipe Anti-inflammatory mechanism in the treatment of rheumatoid arthritis rats.Methods Animal models of CIA rats were established and treated with Qingre Huoxue Fang and honeysuckle.Compare and observe the overall status and morphological changes of each group of rats;pathological changes;ELISA method was used to detect IL-1?and IL-18 in rat serum;RT-PCR was used to detect NLRP3 mRNA expression in rat synovial tissue and PBMC Level;the Western Blot method was used to compare the expression of NLRP3 protein in synovial tissue and PBMC of each group of rats.Results Qingre Huoxue Fang can improve the overall disease status of CIA rats,and the swelling of joints in CIA rats is effectively alleviated.After intervention of Qingre Huoxue Fang,the level of arthritis and synovial tissue proliferation are controlled,and joint bones and cartilage are relieved Damaged;Cytokine results showed:Compared with the model group rats,the contents of IL-18 and IL-1? in the serum of Qingre Huoxue Fang group were reduced(P<0.01,P<0.05);NLRP3 mRNA test results showed:synovial membrane In tissues and PBMC,the levels of NLRP3 mRNA expression in the Qingre Huoxue Fang group and the honeysuckle group were lower than those in the model group(P<0.05).The results of NLRP3 protein detection showed that compared with the synovial tissue of the model group,The expression levels of NLRP3 protein in the formula group and the honeysuckle group decreased(P<0.001).Compared with PBMC in model group,the expression level of NLRP3 protein in Qingre Huoxue Fang group was decreased(P<0.05).Compared with the model group rats,the expression level of NLRP3 protein in the honeysuckle group was reduced,but it was not statistically significant(P>0.05).Conclusions 1.The expression levels of NLRP3 and its downstream factors IL-1? and IL-18 in CIA rats are higher,suggesting that NLRP3 and its downstream factors play an important role in the development of RA.2.Qingre Huoxue Fang group can inhibit the expression of NLRP3 in synovial tissue and PBMC,reduce the content of downstream IL-1? and IL-18 and exert anti-inflammatory effects.NLRP3 inflammatory bodies are expected to become new targets for RA treatment.
