| Objective: To investigate the mechanism of acupuncture in improving the signaling pathway of PCPA insomnia after central 5-HT sleep transmitter and5-HT subtype receptor.Methods: A total of 70 SPF male SD rats were randomly selected from 16 in normal groups,the remaining 54 were made and screened,the patients were randomly divided into model group,acupuncture group(navel inner ring acupuncture point combined with insomnia acupoint prescription)group,non-acupoint group,16 in each group,and normal group,with a total of four groups.The acupuncture group was given acupuncture treatment of internal umbilical ring points and insomnia points by zhuang medicine.Non-acupoint group was given acupuncture of 2 fixed points on each side of the subcostal.No treatment was given in the normal group and the model group,only the same time of scratch stimulation was given.After 6 days of intervention,rat hippocampi were removed.Samples were numbered and saved for inspection accordingly.Morphological changes of 5-HT1 A and 5-HT2 A receptors were detected by immunohistochemistry.The contents of 5-HT in each group were determined by elisa,and AC activity was measured by radioimmunoassay.Use statistical methods to analyze the correlation between 5-HT and AC.Results: 1.Observation of immunomorphology: Compared with the normal group,the number of 5-HT1 AR positive expression in the hippocampus of the model group rats decreased,and scattered and looselyarranged.The number of 5-HT2 AR positive particles increased significantly,and positive expression regions were concentrated.After acupuncture treatments,5-HT1 AR positive cells of acupuncture group became more granular and regular,and the color was darkened.At the same time,5-HT2 AR brown expression significantly reduced,and its positive areas were lighter.2.The experimental indicators showed: A?Compared with the normal group,the expression of 5-HT1 AR in the hippocampus of the model group was lower(P < 0.01),but 5-HT2 AR expression rose(P < 0.01);The content of5-HT was significantly decreased(P < 0.01),and AC activity was reduced(P< 0.01).Meanwhile,there was a positive correlation between 5-HT content damage and AC activity damage in model rats,r=0.648,P < 0.01;B?There was no significant difference between the data of the non-acupoint group and the model group(P > 0.05).Besides,there was not a obvious correlation between 5-HT content and AC activity in non-acupoint rats;C?Compared with the model group,the expression of 5-HT1 AR in the hippocampus of the acupuncture group rose(P < 0.01),but 5-HT2 AR expression was lower(P <0.01);The content of 5-HT increased(P < 0.01),and AC activity increased(P< 0.01).Acupuncture also showed a positive correlation between 5-HT content and increased AC activity,r=0.648,P < 0.01;D ? Compared with non-acupoint group,the expression of 5-HT1 AR of the acupuncture group rose(P < 0.01),but 5-HT2 AR expression was lower(P < 0.01);The content of 5-HT increased(P < 0.01),and AC activity also increased(P < 0.01).Conclusions: 1.PCPA model rats hippocampus show abnormal 5-HT1 A and 5-HT2 A receptor morphology;Acupuncture improve morphology of5-HT1 A and 5-HT2 A receptors in hippocampus of PCPA insomnia model rats.2.The hippocampus of PCPA model rats show 5-HT content and decreased AC activity,and the damage is positively correlated.And acupuncture in the umbilical inner ring and insomnia prescription can significantly increase its5-HT content and up-regulate its AC activity;There is also a correlation between the improvement of the two.3.The experiment support this acupuncture method by improving the 5-HT sleep transmitter,5-HT subtype receptor signaling pathway and impacting interaction mechanism play a role in the treatment of PCPA insomnia. |
