The Effect Of CADM1 On Proliferation And Migration Of Endometrial Carcinoma Cell

Purpose:To investigate the effects of CADM1 overexpression on the proliferation and migration of endometrial cancer cell line(HEC-1-b).And investigate the relationship between the high expression of CADM1 and MMP-9.Methods:Using the lipofection to Transfecting pcDNA3.1-CADM1 eukaryotic expression vector into endometrial cancer cell line HEC-1-b?Using the QRT-PCR to detect the expression of CADM1 mRNA ?Using the Western blotting to detect the expression of CADM1 protein and the expression of MMP-9 protein in endometrial cancer cell line HEC-1-b with high expression CADM1,Using CCK8 method and plate clone formation method to test cell proliferation level and Cell scratch test to detect cell migration ability.Results : 1.Successfully transfected CADM1 overexpression into endometrial cancer cells HEC-1-b;and with the expression of CADM1 increased,the expression of MMP-9 was decreased(P <0.01).2.Compared with the empty vector group and the blank control group,the CADM1 overexpression group had reduced proliferationactivity of the endometrial cancer cell HEC-1-b(P <0.01),decreased the number of cell clones formed(P <0.01),and decreased the migration ability(P <0.01).).It shows that high expression of CADM1 can inhibit the proliferation and migration of endometrial cancer.Conclusion : 1.High expression of CADM1 can inhibit the proliferation and migration ability of endometrial cancer cell.2.High expression of CADM1 inhibited the expression of MMP-9.
