Objective:We investigated whether leptin mediates the protection effect of H2S with the expression of ALDH2 and ADH1C in the cerebral cortex of 6-OHDA-induced Parkinson disease.Methods:One hundred SD rats were choosen to adapt to surrounding within one week,and then SD rats were divided into seven groups:control group,PD group,PD+NaHS?30?mol/kg/d,i.p.?group,PD+NaHS?100?mol/kg/d,i.p.?group,PD+NaHS+leptin-OBR?2 ug,i.c.v.?group,NaHS?100?mol/kg/d,i.p.?group,leptin-OBR?2 ug,i.c.v.?group.The expressions of ALDH2 and ADH1C in the cerebral cortext were detected by Western blotting.The data was analyzed by SPSS20.0.Results:1.H2S promote the expression of ALDH2 in the 6-OHDA-treated rats.The expression of ALDH2 in the cerebral cortext of PD group decreased compared with control group?P<0.05?.But the expression of ALDH2 in the cerebral cortext of PD+NaHS?100?mol/kg/d,i.p.?group increased compared with in the cerebral cortext of PD group?P<0.05?.These results indicate that H2S antagonist the decrease in the expression of ALDH2.2.H2S promote the expression of ADH1C in the 6-OHDA-treated rats.The expression of ADH1C in the cerebral cortext of PD group decreased compared with in the cerebral cortext of control group?P<0.01?.But the expression of ADH1C in the cerebral cortext of PD+NaHS?100?mol/kg/d,i.p.?group increased compared with in the cerebral cortext of PD group?P<0.05?.These results indicate that H2S antagonist the decrease in the expression of ADH1C.3.H2S promote the expression of ALDH2 in the 6-OHDA-treated rats by upregulation of leptin.The expression of ALDH2 in the cerebral cortext of PD+NaHS+leptin-OBR?2 ug,i.c.v.?group decreased compared with in the cerebral cortext of PD+NaHS?100?mol/kg/d,i.p.?group?P<0.001?.These results indicate that H2S antagonist the decrease in the expression of ALDH2 by upregulation of leptin.4.H2S promote the expression of ADH1C in the 6-OHDA-treated rats by upregulation of leptin.The expression of ADH1C in the cerebral cortext of PD+NaHS+leptin-OBR?2 ug,i.c.v.?group decreased compared with in the cerebral cortext of PD+NaHS?100?mol/kg/d,i.p.?group?P<0.01?.These results indicate that H2S antagonist the decrease in the expression of ADH1C by upregulation of leptin.Conclusion:Leptin mediated hydrogen sulfide antagonist the decrease in expressions of ALDH2 and ADH1C in the cerebral cortext of Parkinson's disease rats. |
