[Background and Objective]Parkinson's disease?PD?is a neurodegenerative disease characterized by degeneration and necrosis of the middle cerebral striatum dopaminergic neurons.Hydrogen sulfide?H2S?acts as a novel gas signaling molecule with neuroprotective effects.The development of PD to synaptic plasticity in the late stage of the disease will explore whether H2S can improve hippocampal synaptic plasticity in PD rats,in order to provide a new method for the treatment of PD patients.[Methods]1.Rotenone?ROT?was injected into the rat to establish a PD model;2.Western blot?WB?detected the expression of hippocampal synaptic plasticity-associated protein,including synapsin-1?Syn-1?and post-synaptic density protein-95?PSD-95?;3.Transmission electron microscopy observed the changes of hippocampal synaptic microstructure and density.[Results]1.H2S can increase the expressions of hippocampal synaptic-associated proteins in PD rats:WB results showed that the expressions of synaptic-associated proteins Syn-1 and PSD-95 in hippocampus of PD rats were significantly lower than those in the control group.NaHS significantly increased the expressions of synaptic-related proteins in the hippocampus of Syn-1 and PSD-95in the hippocampus of PD rats.2.H2S can ameliorate synaptic density in hippocampal CA1 region of PD rats:The synaptic microstructure of hippocampal CA1 region was detected by transmission electron microscopy.The synaptic density in hippocampal CA1 region of PD rats was significantly decreased in PD rats,while NaHS remarkablely increased.3.H2S can improve the synaptic interface structure in hippocampal CA1 region of PD rats:The synaptic microstructure of rat hippocampal CA1 region was detected by transmission electron microscopy.The length of synaptic activity area and post-synaptic density of PD rats were decrased,the synaptic gap was increased,in the hippocampus?CA1?of PD rats.NaHS significantly increased the length of synaptic activity area and The density of post-synaptic density and the synaptic gap width decreased in the hippocampus?CA1?of PD rats.[Conclusion]H2S can improve synaptic plasticity in the hippocampus of PD rats.
