Objective:Lipid metabolism is a major factor for atherosclerosis(AS).From the perspective of regulating cholesterol metabolism in ApoE-/-mice,the mechanism of spleen-reducing turbidity classic prescription Zexieyin on the prevention and treatment of atherosclerosis was provided to provide effective theoretical basis for its clinical application.Method:Forty male 8-week-old ApoE-/-mice were randomly divided into model group(HF D),low-dose ZXY group(LZXY).high-dose group(HZXY),and atorvastatin group(Ato).10 mice in each group,and 10 C57BL/6J mice served as a blank control group(Con).Contr of group was given normal diet.Model group.low-dose ZXY group,high-dose group,ator--vastatin group,were given high-fat diet.At the same time,low-dose ZXY group,high-dose group,atorvastatin group were intragastrically administrated with corresponding drugs.After 9 weeks,triglyceride(TG),cholesterol(TC),low-density lipoprotein cholesterol(LDL-C)a nd high-density lipoprotein cholesterol(HDL-C)were detected.Oil red "O" staining method was used to observe the pathological changes of the aortic arch root,and the ratio of lesion area in the lumen was determined and analyzed.Western blot was used to determine the pr otein expression levels of LXRa,CYP7A1,ABCG5,ABCG8 in the liver tissue and LXR?.SREBP-2,NPCIL1.ABCG5,ABCG8 in the small intestine tissue.Result:In the terms of serum lipid levels,compared with the control group,the levels of T C,TG,LDL-C,and HDL-C in the ApoE-/-mice model group were significantly increased(P<0.01);compared with the model group,HZXY Group,LZXY group and atorvastatin gro up can all reduce TC and TG levels in ApoE-/-mice(P<0.01);the level of LDL-C in HZX Y group and atorvastatin group was Significantly decreased(P<0.01),but the change of H DL-C level was not statistically significant.Pathological morphological observation showed t hat compared with the control group,the intima of the aorta in the model group was thicke ned and raised,lipid droplets were accumulated,and there were a large number of plaques Block formation;compared with the model group,HZXY group,LZXY group,and atorvast atin group can effectively decrease the proportion of plaque area in the aortic lumen(P<0.01).Cholesterol transport and metabolism-related protein levels,compared with the model g roup,the HZXY group and the atorvastatin group significantly increased the expression leve I of CYP7A1 protein in liver tissues(P<0.01).The HZXY group expression levels of ABC G5,ABCG8 and LXR? in liver were significantly increased(P<0.01);the expression levels of ABCG8 protein in liver in the LZXY group were significantly increased(P<0.05).Com pared with the model group,HZXY group and LZXY group significantly increased LXR? p rotein expression in the small intestine(P<0.05);and HZXY group significantly increased t he intestine protein expression level of ABCG5,ABCG8 and decreased the protein expression of NPCIL1 and SREBP2 in the small intestine.Conclusion:Zexieyin has a preventive effect on the occurrence and development of AS,an d its mechanism may be related to regulating cholesterol metabolism,inhibiting the absorpti on of cholesterol in the intestine,and promoting the excretion of cholesterol.
