Objective: To detect the expression level of annexinA3(ANXA3)mRNA and its protein in breast cancer tissues and normal tissues adjacent to cancer,and analyze the correlations between annexinA3 and clinicopathological parameters of breast cancer patients,to discuss the clinical significance of annexinA3 in breast cancer ulteriorly.Methods: We randomly selected cancer tissues and normal tissues adjacent to cancer of 50 breast cancer patients who were pathologically diagnosed as primary breast cancer from the department of breast surgery,the Second People's Hospital of Yibin,Sichuan Province,from March 2019 to May 2019.All cases were required to have complete information and receive no treatment before operation.The relative expression level of ANXA3 mRNA and its protein in breast cancer tissues(experimental group)and the normal tissues adjacent to cancer(control group),both of which were from the same patient,were determined by real-time fluorescence quantitative polymerase chain reaction(FQ-PCR)and Western Blot respectively,to compare the expression level of ANXA3 in the cancer tissues and normal tissues adjacent to cancer.the association between the expression of ANXA3 mRNA and its protein in breast cancer tissue and normal tissue adjacent to cancer was analyzed by Pearson product moment correlation coefficient.Finally,the experimental group was divided into different subgroups by different clinicopathological parameters(including age,menstrual conditions,tumor size,lymph node metastasis,TNM stage,molecular subtypes,histological grade,nerve invasion,and vascular invasion).Further analysis of the correlations between ANXA3 mRNA and its protein and the clinicopathological parameters of breast cancer patients.The data obtained in this experiment was analyzed by SPSS 24.0,and the role of ANXA3 in the genesis,development and metastasis of breast cancer was furtherly discussed.Results: 1.The expression level of ANXA3 mRNA in breast cancer(4.2538±2.3403)were apparently higher than that in normal tissues adjacent to cancer(1.0082±0.4730),and the difference was statistically significant(t=10.221,P<0.001).The expression level of ANXA3 protein in breast cancer(1.1234±0.6193)were apparently higher than that in normal tissues adjacent to cancer(0.6418±0.3215),and the difference was statistically significant(t=5.733,P<0.001).2.The result of Pearson product moment correlation coefficient showed that there was a roughly moderate positive correlation between expression of ANXA3 mRNA and its protein in breast cancer tissues and a normal tissues adjacent to cancer(P<0.001).3.The patients were divided into different subgroups by different clinicopathological parameters.The results showed that the relative expression of ANXA3 mRNA and its protein were not significantly correlated with age,menstrual conditions,tumor size,TNM stage and histological grade(P>0.05),but with lymph node metastasis and molecular subtypes(P<0.001).The relative expression of ANXA3 mRNA and its protein in breast cancer with lymph node metastasis were higher than that without lymph node metastasis,and the relative expression level of ANXA3 mRNA and its protein in breast cancer with the number of lymph node metastasis more than 4 were higher than that in breast cancer with the number of lymph node metastasis 1-3(P<0.001).The expression of ANXA3 mRNA and its protein were various among different molecular subtypes,and those were higher in triple negative breast cancer,followed by HER-2 overexpression(P<0.001),there was no significant difference between Luminal A and Luminal B(P>0.05).Conclusions: The expression of ANXA3 mRNA and its protein in breast cancer were significantly up-regulated,and the expression in breast cancer with lymph node metastasis were higher than that without lymph node metastasis.The results suggest that ANXA3 may be involved in the process of tumor genesis,development and metastasis,and it is expected to provide available information for the desease of surveillance and prognosis prediction of breast cancer and to provide the direction for finding novel biological markers of breast cancer,and it may had potential value.High expression of ANXA3 in triple negative breast cancer may indicate that ANXA3 is related to triple negative breast cancer and may provide available information for further exploration of biological behavior of triple negative breast cancer.
