| Objective: To investigate the expression level of microRNA-378-3p(miRNA-378-3p)in the tissues of patients with cutaneous squamous cell carcinoma(CSCC)and the effect of the latter on proliferation,apoptosis,invasion and migration of cancer cells.Methods: The tissue samples of 83 CSCC patients hospitalized in hospital from Feb 2017 to Feb 2019 were collected.Using the fluorescent quantitative PCR detection in the tissue adjacent to carcinoma tissues and the expression of the miRNA-378-3p differences.According to the situation of the miRNA-378-3-p transfection CSCC of cell line A-431 can be divided into the miRNA-378-mimics group,the miRNA-378-inhibitor group and blank control group.CCK 8 method was used to detect three groups of cells in 0h,6h,12 h,24h,48 h point proliferation,Annexin v-fitc /PI double staining was used to detect the apoptosis of cells in each group,and Transwell assay was used to detect the migration and invasion ability of cells in the three groups.In addition,the target gene of mirna-378-3p was predicted by Targetscan database,the target gene CD226 related to tumor proliferation and apoptosis was selected to detect the expression level of CD226 in each group by Western Blotting,and the interaction between mirna-378-3p and CD226 was detected by double-lucifase reporting experiment.Results: The expression level of miRNA-378-3p in cancer tissues of CSCC patients was significantly higher than that in adjacent tissues(P<0.001).Compared with the blank control group,the content of CD226 in the miRNA-378-mimics group was lower(P<0.001).The results of the bilucifer enzyme assay showed that CD226 was the downstream target gene of miRNA-378-3p(P<0.001).Compared with the control group and the miRNA-378-inhibitor group,the proliferation rate of cells in the miRNA-378-mimics group was significantly increased while the apoptosis rate was decreased(P<0.001,P=0.02),cell migration and invasion were significantly enhanced(P<0.001,P<0.001).Conclusion: MiRNA-378-3p is up-regulated in cancer tissues and can regulate the proliferation,apoptosis,invasion and migration of cancer cells by targeting CD226,suggesting that miRNA-378-3p can play a role similar to that of oncogenic genes and may be a target for CSCC diagnosis and novel biotherapy. |
