Objective:Exploring Buzhong Yiqi Decoction on micro RNA-21(micro RNA-21,miRNA-)in tumor tissue and normal tissues adjacent to the tumor in patients with pus ear disease(weak spleen and stomach)-Middle Ear Cholesteatoma 21).The effects of programmed cell death 4(PDCD4)and the expression levels of miRNA-21 and PDCD4 in the tumor tissue of the middle ear cholesteatoma rat model,based on the miRNA-21 / PDCD4 signaling pathway To explain the mechanism of Buzhong Yiqi Decoction in treating the proliferation and apoptosis phenotype of middle ear cholesteatoma(MEC)cells.Methods: A randomized,controlled,prospective study was used.(1)Collect 10 postoperative tumors and normal tissue samples of patients with spleen and stomach weak type pus ears who were hospitalized in the Department of Otorhinolaryngology of our hospital from June 2019 to December 2019 and received middle ear cholesteatoma removal surgery.Quantitative Real-time PCR(q Rt-PCR)method(the housekeeping gene is ?-actin)was used to detect the expression level of miRNA-21 in each tissue,and Western blot(WB)was used to detect the expression of PDCD4.;(2)At the same time,select healthy rats weighing 200 ± 25 g,and exclude eardrum perforation and middle ear infection through ear microscopy,and use middleear chemical reagent(propylene glycol)injection method to establish middle ear cholesteatoma rats(single Side)The model is eliminated,and the rats that failed to make the model and failed are randomly divided into the experimental group and the control group.The tumor volume of the two groups is measured and recorded.The experimental group is given the Chinese medicine Buzhong Yiqi Decoction with a dose of 2.67 g / kg Stomach,once a day in the morning,continuous gavage for 4 weeks;the control group was gavaged with the same amount of normal saline;during the period,the tumor was observed through the microscope.After the last gavage,the rat was killed by cervical decompression.Remove the tumor Drainage using Archimedes' principle,measured with a graduated cylinder and record the volume of the tumor in vitro.After the removed tumor tissue samples were fully lysed on ice,the expression of miRNA-21 in the tissue was analyzed by real-time fluorescence quantitative PCR,and the expression level of PDCD4 in the tissue was detected by Western blot.The experimental data obtained was sorted out,and the quantitative data was expressed as mean ± standard deviation.SPSS22.0 software was used for data statistics and analysis.The data between the two groups were paired t-test,with P <0.05 as the standard to indicate that there is difference Learn meaning.Results:Human specimen test part:(1)Compared with the expression level of miRNA-21 in normal tissues adjacent to the tumor,the expression level of miRNA-21 in tumor tissues is high,and the difference between the two groups is statistically significant(P <0.05),(2)The expression level of PDCD4 in tumor tissues was significantly lower than that in normal tissues adjacent to the tumor,and the difference was statistically significant(P <0.05);the experimental part of rats:(3)Jingbuzhongyi After Qitang intervention,the tumor volume of the experimental group was smaller than that before the intervention,and there was a significant difference(P <0.05).(4)The expression level of miRNA-21 in the tumor tissue of the experimental group was lower than that of the control The difference between the two groups was statistically significant(P <0.05).(5)The expression level of PDCD4 in the tumor tissue of the experimental group was significantly higher than that of the control group,and the difference was statistically significant(P <0.05).Conclusion:miRNA-21 and PDCD4 are key regulatory targets for the occurrence and development of pustulosis(middle ear cholesteatoma).miRNA-21 / PDCD4 signaling pathway is closely related to the pathogenesis of middle ear cholesteatoma Lipoma,by strengthening the spleen and replenishing qi,is used to correct the proliferative phenotype of the disease by down-regulating miRNA-21 and up-regulating the PDCD4 signaling pathway.
