Objective: The purpose of this study was to investigate the therapeutic effect of growth hormone on thin endometrium in rats.Method: Forty-eight 7-week-old female SD rats were randomly divided into blank control group,model group,subcutaneous group and growth hormone group.The blank control group was not subjected to any operation and was routinely reared.Vaginal smears were performed in the remaining groups at 9 am and 5 pm daily,and female rats with a complete estrous cycle were tested.Rats' uterus was perfused with 95% ethanol in the model,subcutaneous and growth hormone groups to establish a thin endometrium model.After model surgery,6-8 hours later,the model group was perfused intrauterinely with 0.2 ml of normal saline,the subcutaneous group was injected subcutaneously with 0.2 ml of growth hormone,and the growth hormone group was perfused with 0.2 ml of growth hormone.After the rats were rested for two weeks,the model group,subcutaneous group,and growth hormone group were subjected to vaginal smears at 9 a.m.and 5 p.m.each day.The rats were sacrificed during the estrus period.The uterus of each rat and rat was stored at-80?,and partly fixed at 10% neutral formalin.After paraffin fixation,relevant experimental verification was performed.After paraffin sections,hematoxylin and eosin staining were used to measure endometrial thickness.Immunohistochemistry was used to detect the expression of cytokeratin and vimentin in rat endometrial tissue.Immunoblotting was used to detect the expression of cytokeratin and vimentin in rat endometrial tissue.Results: The intimal thickness of the growth hormone group was greater than that of the model group(P <0.01),the intimal thickness of the blank control group was greater than that of the model group(P <0.01),and the intimal thickness of the subcutaneous group was greater than that of the model group,but the difference was not statistically significant.The number of uterine blood vessels in the growth hormone group was greater than that in the model group(P <0.01),the number of uterine blood vessels in the blank control group was greater than that in the model group(P <0.01),the number of glands in the growth hormone group was greater than in the model group(P <0.01),and the glands in the blank control group was greater than in the model group(P <0.01).The number of uterine blood vessels in the subcutaneous group was greater than in the model group,but the difference was not statistically significant.The number of uterine glands in the subcutaneous group was greater than in the model group,but the difference was not statistically significant.The expression of vimentin in the growth hormone group(P <0.01)was higher than that in the model group(P <0.01),and the expression of cytokeratin in the growth hormone group(P <0.01)was higher than that in the model group(P <0.01).Conclusion: Intrauterine injection of growth hormone can treat rat thin endometrium,intrauterine infusion of growth hormone can promote the regeneration and repair of rat thin endometrium;Intrauterine injection of growth hormone may be better than subcutaneous injection of growth hormone in the treatment of rat thin uterus.
