Objective:To study the mechanism of SDF-1/CXCR4 signal axis regulating Knee oateoarthritis(KOA)pain induced by abnormal mechanical stress and the intervention effect of"Xibining"" in this link.Method:1.The safranin O staining?Mankin grade score?and the mRNA expression of MMP-13 and Co12al in the cartilage were used to evaluate the pathological changes of KOA.Bilateral balance pain meter was used to detect lower limb pain in rats and western blot was used to detect related indicators in subchondral bone,including TRAP?SDF-1?CXCR4?netrin1?NGF.VEGF mRNA expression in subchondral bone was detected by RT-qPCR to observe the change of angiogenesis activity.2.To observe the effect of mechanical stress on SDF-1/CXCR4 signaling pathway of osteoclast and the intervention effect of "Xibining",the mRNA and protein expression of SDF-1?netrin1 and NGF were detected by RT-qPCR and ELISA.3.To observe the effect of SDF-1 on osteoclast and the intervention effect of "Xibining",the mRNA and protein expression of SDF-1 and NGF were detected by RT-qPCR and ELISA.Results:1.Protein expression of TRAP?SDF-1?CXCR4?netrin1?NGF in subchondral bone of "Xibining" group were significantly decreased compared to KOA group,the difference was significant(p<0.05).The load-bearing ratio of knee joint in"Xibining" group were significantly up-regulated compared to KOA group(p<0.05).The mRNA expression of MMP-13 was decreased and the mRNA expression of Co12al was increased in "Xibining" group compared to KOA group(p<0.05).The Mankin score was significantly decreased in "Xibining" group compared to KOA group(p<0.05).2.The mRNA expression of SDF-1?netrin1?NGF?TRAP in abnormal mechanical stress group was significantly increased compared to normal group(p<0.05)and 5%mechanical stress group was the most obvious.The mRNA expression of SDF-1?netrin1?NGF?TRAP were significantly decreased after intervention with drug serum of "Xibining".3.The mRNA expression of CXCR4?netrin1?NGF in osteoclast were increased in SDF-1 group compared to normal group(P<0.05).The mRNA expression of CXCR4,netrinl,NGF in osteoclast were decreased in AMD3100 group and "Xibining"group compared to SDF-1 group(P<0.05).There was no difference in the mRNA expression of CXCR4?netrin1?NGF between AMD3100 group and "Xibining"group(p>0.05).Conclusion:1.The protein expression of TRAP?SDF-1?netrin1?NGF in subchondral bone were significantly reduced by "Xibining",which relieved the KOA pain in KOA.2.The increased mRNA expression of SDF-1?netrin1?NGF in osteoclast induced by abnormal mechanical stress were significantly inhibited by"Xibining".3.Osteoclast promoted the mRNA expression of SDF-1?netrin1?NGF by the activation of SDF-1/CXCR4 pathway signal,"Xibining" significantly inhibited the activation of SDF-1/CXCR4 pathway signal in osteoclast.
