In recent years,the incidence of obesity has increased rapidly,and the problem of obesity has become increasingly severe,which has brought tremendous pressure and burden to people's lives and economy.Drug treatment of obesity may have certain side effects.Surgical treatment of obesity also has certain risks,and it will also cause certain negative emotions to people's psychology.At present,scientific diet and proper exercise are the most effective health methods for maintaining and reducing body weight.Studies have shown that probiotics are a group of microorganisms that can be colonized in a certain amount in the human intestine to exert various physiological effects on the body,and can prevent and relieve certain diseases to a certain extent.Therefore,foods containing probiotics are widely concerned,such as beverages and dietary supplements.This thesis takes C57BL/6J mice as the research object and uses a high-fat diet-induced obesity model to explore Lactobacillus casei NCU011054,Lactobacillus fermentum NCU0413,Lactobacillus acidophilus NCU433,Lactobacillus rhamnosus NCU2217,Lactobacillus paracasei NCU622 Intervention effect on obese mice induced by high-fat diet.The main research contents and conclusions are as follows:(1)45%high-fat diet D12451(Research Diets Inc.)was used to induce obesity in male C57BL/6J mice.After 9 weeks,the model was successfully constructed.After the intervention of five strains of Lactobacillus,the results showed that the weight growth trend of obese mice was well suppressed,and the weight gain was significantly reduced,the Lee's index was reduced,the body fat and body fat rate were significantly reduced(p<0.05),and fasting blood glucose was also significantly reduced in mice.After 9 weeks of five Lactobacillus interventions in obese mice induced by a high-fat diet,serum TC,TG,and LDL in obese mice were significantly reduced(p<0.05),and HDL was significantly increased(p<0.05).It shows that these five strains of Lactobacillus can effectively regulate the disorder of serum lipids and cholesterol.At the same time,the serum leptin and adiponectin levels of the mice in the LC and LA groups were measured.The results show that after the intervention of Lactobacillus,the LEP content in the serum of the LC low,middle and high dose group was significantly lower than that of the HFD group(p<0.05).The serum LEP content in the LAM group was lower than that in the HFD group,but it was not significant,while the LAL and LAH groups were significantly lower than those in the HFD group(p<0.05).In addition,the serum ADPN levels of mice in the HFD group were lower than those in the other groups,and significantly lower than those in the LCH,LAL,LAM,and LAH groups(p<0.05).The above results show that the two Lactobacillus strains,LC and LA,have a certain regulation effect on serum leptin and adiponectin levels in obese mice.The above results indicate that these five strains of lactobacillus have anti-obesity effects.(2)To investigate the effects of five strains of Lactobacillus on functional tissues of obese mice,the basic indexes of liver function and kidney function were measured.Through the determination of the basic indexes of liver function and kidney function,the five strains of Lactobacillus have a reducing effect on liver and kidney indexes,and have a regulating effect on ALT,AST,CREA-J,UA,etc.This indicates that five strains of Lactobacillus have better regulation of liver and kidney dysfunction caused by a high-fat diet,and the effect of LP is better than the other four strains of Lactobacillus,In addition,five strains of Lactobacilli can improve liver enlargement in obese mice induced by a high-fat diet.Besides,from the results of HE staining of the liver,it can be seen that these five strains of Lactobacilli can improve fatty liver lesions caused by long-term high-fat diet.And LP is the best and LF is the least.According to the results of TC and TG content in the liver of obese mice,the five strains of Lactobacillus can significantly reduce the levels of TC and TG in the liver of obese mice with high-fat diet,and the effect of lowering TG is better.Lactobacillus inhibits the accumulation of lipids in the liver.And the effect of Lactobacillus in LC group is average,and the effect of LA is the best.By measuring the antioxidant capacity of mouse liver,these five strains of Lactobacillus can increase the levels of liver antioxidant enzymes GSH-Px,T-SOD,and CAT,which has a better promotion effect on the body's elimination of excessive free radicals.In addition,with the intervention of five lactic acid bacteria,liver MDA levels decreased and T-AOC levels increased.These five strains of Lactobacillus have good antioxidant effects and can inhibit oxidative damage to the liver caused by obesity.Observation of pathological sections of adipose tissue shows that the fat cells in the LCM,LCH,LFH,LAM,LAH,LRM,LRH,LPM,and LPH groups are neatly arranged,a small number of cells are large in size and appear adipose differentiation,and the overall cell size is similar to that of the ND group,and even the number of cells in the same field is smaller than that of the ND group.The contents of pro-inflammatory factors LEP,TNF-?,IL-6,IL-1? and anti-inflammatory factors ADPN and IL-10 in the adipose tissue of mice in the LR and LP groups were also determined.The results showed that LR and LP strains had a certain regulation effect on the pro-inflammatory factors LEP,TNF-?,IL-6,IL-1? and anti-inflammatory factors ADPN and IL-10 in adipose tissue of obese mice.In summary,these five strains of Lactobacillus have an improvement effect on the body tissues of obese mice.(3)Further investigating the intestinal health status of obese mice:HE staining of colon tissue showed that these five strains of lactobacilli can improve the micro-inflammation of the intestine caused by a long-term high-fat diet.Compared with the HFD group,the crypt structure of the colonic mucosa layer in the lactobacillus group was neater,the goblet cells were more complete,and there was basically no inflammatory cell infiltration.This shows to a certain extent that Lactobacillus has a certain protective effect on the intestinal mucosa and has health promotion effect on the intestinal environment,and the LC and LA strains effects are better.Except that the concentrations of SCFAs in the colonic contents of the mice fed the lactobacillus group were higher than those in the LRL group,the acetic acid and propionic acid in the other groups were lower than those in the HFD group;n-butyric acid was slightly lower than in the HFD group.This may be due to the compensatory change of other metabolites in the intestinal environment caused by gavage of Lactobacillus,which indirectly increased the levels of other metabolites in the intestine,which is worthy of our further research.In summary,this paper initially explored the improvement of the symptoms of five strains of Lactobacillus on obese mice induced by high-fat diet.The results show that the five strains of Lactobacillus can better suppress the obesity symptoms in mice.These results will lay a certain preliminary foundation for further research in the future.
