Background and ObjectiveThe incidence of stroke in China is increasing year by year,and it has already become the disease of the highest morbidity in our country.Stroke has the characteristics of high disability and high recurrence,which seriously endangers the life and health of residents and continuously increases the burden of disease in our country.The main effective treatment of acute ischemic stroke is recanalization of blood vessels within the time window at the moment,including interventional mechanical thrombectomy and intravenous thrombolysis.The blood-brain barrier is an important structure to maintain the homeostasis of the central nervous system,and pericytes are one of its main components.Pericytes are widely distributed in the central nervous system,and mainly located in the basement membrane of capillaries.The main roles of pericytes in stroke include regulating cerebral blood.flow,maintaining the blood-brain barrier' stability,and promoting angiogenesis.Previous studies have shown that the subventricular zone is one of the important areas of neurogenesis.After cerebral infarction,neurogenesis is enhanced in SVZ,and newborn neuroblasts can migrate,integrate and differentiate into the infarcted area to rebuild the neural network around the infarcted areas,which can promote the recovery of neurological function.Resveratrol is a non-flavonoid polyphenol compound,which has several biological activities,such as anti-inflammation,anti-oxidation,and anti-apoptosis.In this study,we firstly prepare the model of mice cerebral infarction,and then resveratrol was administered to observe vascular endothelial growth factor B,pericytes,and the recovery of neurological function in mice brain.The purpose of this study is to investigate the effect of resveratrol on the survival of pericytes around the infarction areas and the neurogenesis in subventricular zone.MethodsAdult Kunming mice were randomly divided into the sham+vehicle group,the cerebral infarction+vehicle,and the cerebral infarction +Tresveratrol group.The transient middle cerebral artery occlusion model was made by the suture method,and the amount of VEGFB was determined by western blot.Pericytes were labeled with PDGFR?,proliferative cells were labeled with BrdU,and neuroblasts were marked with DCX,then the number of pericytes around infarction areas and neuroblasts in SVZ of every group of mice was detected by immunofluorescence.Finally,we use mNSS to assess mouse behavior.Results1.After cerebral infarction,the brain water content and modified neurological severity scores of mice was significantly increased,and the number of pericytes around infarction areas was obviously decreased,but the number of neuroblasts in SVZ was significantly increased.2.Compared with the cerebral infarction+vehicle group,the brain water content and modified neurological severity scores of mice were decreased,and the number of pericytes around infarction areas and neuroblasts in SVZ of mice were obviously increased of the cerebral infarction+resveratrol group.3.After cerebral infarction,the expression of VEGFB was raised.Compared with the cerebral infarction+vehicle group,the expression of VEGFB from the mice of the cerebral infarction+resveratrol group was higher.ConclusionsResveratrol may promote the survival of pericytes around the infarction areas and neurogenesis in SVZ by partially up-regulating VEGFB.
