Hepatocellular carcinoma(HCC)is the second fatality rate malignancy tumor worldwide.The onset of HCC is insidious,and most cases are found in the middle or late stages.Although surgical resection is the most effective treatment for liver cancer,many patients have lost their chance of surgery when discovered.Currently,the clinical effect of chemotherapy on liver cancer is limited,and patients with liver cancer are prone to develop resistance to chemotherapy drugs.Therefore,in recent years,the search for specific markers of liver cancer and the exploration of molecular targeted therapy for liver cancer has become a research hotspot.STMN1,a small-molecule protein with a molecular weight of 19kDa,has been widely studied in recent years.It is expressed in most normal tissues and overexpressed in a variety of tumor tissues.It can change microtubule dynamics during mitosis and intermitosis and participate in cell proliferation,movement and cycle regulation.STMN1 is expected to be a new target for the diagnosis and treatment of malignant tumors.Currently,various studies on the relationship between STMN1 and malignant tumors are being actively carried out,but there are few studies on the relationship between STMN1 and hepatocellular carcinoma.Therefore,this topic will study the relationship between STMN1 and hepatocellular carcinoma from the following two aspects:1.The expression of STMN1 in hepatocellular carcinoma and para-carcinoma was detected by immunohistochemistry(IHC)and Western Blot methods.Respectively,The relationship between the expression of STMN1 in hepatocellular carcinoma and the clinical characteristics and prognosis of patients was analyzed.2.Silencing the expression of STMN1 gene in HepG2 cells using small interfering RNA(siRNA).Western Blot method was used to test the silencing efficiency of siRNA on STMN1.MTT method was used to detect cell viability,Annexin V-FITC/PI double staining method was used to detect apoptosis,and the relationship between STMN1 and hepatocellular carcinoma was studied at the cell level.Part?:Expression of STMN1 in Hepatocellular Carcinoma and its Clinical SignificanceObjectiveImmunohistochemistry(IHC)and Western Blot were used to detect the expression of STMN1 in hepatocellular carcinoma and adjacent tissues.The relationship between the expression of STMN1 in liver cancer tissues and the clinical characteristics and prognosis of patients was analyzed.Methods1.Western Blot method was used to detect the expression of STMN1 in fresh samples of 20 hepatocellular carcinoma tissues and corresponding paracancer tissues.2.Immunohistochemical method was used to detect the expression of STMNI in 90 cases of hepatocellular carcinoma tissues and corresponding paraffin tissues.3.To analyze and compare the expression of STMN1 in hepatocellular carcinoma and paracancer tissues,and to explore the relationship between the expression of STMN1 and the clinical characteristics of HCC patients.4.Survival analysis explored the relationship between the expression of STMN1 and the prognosis of HCC patients.Results1.The results of immunohistochemistry and Western Blot showed that the expression level of STMN1 in HCC tissues was significantly higher than that in para-carcinoma tissues(P<0.05).2.Statistical analysis of clinical characteristics showed that the expression of STMN1 in HCC was independent of the age,gender and tumor size of the patients(P>0.05),and correlated with TNM stage and differentiation degree(P<0.05).3.The expression of STMN1 in hepatocellular carcinoma was negatively correlated with the survival time and the overall survival time(P<0.05).Conclusion1.The expression of STMN1 in hepatocellular carcinoma tissues was significantly higher than that in adjacent tissues.2.The expression of STMN1 was not related to the age,gender and tumor size of HCC patients,was related to TNM stage and differentiation degree,and was negatively correlated with the tumor-free survival time and overall survival.3.The high expression of STMN1 may have an important effect on the occurrence,development and prognosis of hepatocellular carcinoma.Part?:Effects of Silencing STMN1 Expression in HepG2 Cells on Proliferation and ApoptosisObjectiveSmall interfering RNA(siRNA)was used to silence the expression of STMN1 in HepG2 cells,and the effect of low STMN1 expression on cell biological behavior was observed.Method1.HepG2 cells were transiently transfected with siRNAs targeting STMN1 and siRNAs targeting unrelated sequences.Western Bolt method was used to detect the expression of STMN1 in the cells after transfection.2.MTT method was used to detect cell viability after transfection.Annexin V-FETC/PI double staining method was used to detect cell apoptosis rate after transfection on flow cytometer.Result1.The expression level of STMN1 in HepG2 cells was significantly reduced after transfection with small interfering RNA targeting STMN1(STMN1-siRNA)(P<0.05).2.Transfection of HepG2 cells with small interfering RNA targeting STMN1(STMN1-siRNA)significantly reduced cell viability compared with the control group(P<0.05)and significantly increased the apoptosis rate compared with the control group(P<0.05).Conclusion1.STMN1 targeting small interfering RNA transfection can significantly reduce the expression of STMN1 in HepG2 cells.2.After transfection of HepG2 cells with small interfering RNA targeting STMN1,the cell viability was significantly reduced and the apoptosis rate was significantly increased.STMN1 could affect the biological function of liver cancer cells.
