Objective: To study the model of alcohol-induced fatty liver and the mechanism of Paeonol in the treatment of alcoholic fatty liver,and to clarify the role of the Adip/CaMKK?/AMPK signaling pathway in the treatment of AFLD by Paeonol.Method: 1.Firstly,the liver pathological sections were used to verify whether the alcohol-induced fatty liver model in mice was established.Secondly,based on the AFLD model,the levels of inflammatory factors IL-6,IL-1? and TNF-? and the levels of TG and TC in the serum of mice were detected by ELISA.Meanwhile,the nucleic acid levels of AdipR1,CaMKK?,AMPK?,and SREBP1 c in mouse liver tissues were detected by semi-quantitative PCR and qRT-PCR.The protein levels of AdipR1?P-CaMKK??CaMKK??P-AMPK??AMPK? and SREBP1 c in mouse liver were detected by Western blot;The expression of Adip and ACC was detected by ELISA.2.AML-12 cells were induced with alcohol,and the Oil Red O staining method was used to verify the AFLD model of the cells.Based on the AFLD model,the levels of inflammatory factors and TG and TC in AML-12 cells were detected by ELISA.At the same time,semi-quantitative PCR method,qRT-PCR method was used to detect nucleic acid level,and Western blot method and ELISA method were used to detect protein expression level in the above animal experiments.Results: Regardless of the mouse in vivo or in vitro cell line,compared with the control group,under the induction of alcohol,Adip level was significantly reduced(P<0.01),but AdipR1 expression level was not significantly changed(P>0.05);IL-1?,TNF-? and TG,TC contents were significantly increased(P<0.01);Both nucleic acid level(P<0.05)and protein phosphorylation level of CaMKK? and AMPK? decreased significantly(P<0.01);the expression of SREBP1 c increased significantly(P<0.01),and the content of ACC also increased significantly(P<0.01).Compared with Et OH group,Pae group of Adip levels was increased(P<0.05),but its receptor 1 remained unchanged(P>0.05),the expression of inflammatory factors was decreased(P<0.05),and the fat content(TG,TC)was significantly reduced(P<0.05).For CaMKK? and AMPK?,there was a significant increase in nucleic acid level and protein phosphorylation level(P<0.01);SREBP1c expression was inhibited(P<0.05)and ACC content was significantly reduced(P<0.05).Conclusions: The mechanism of paeonol in the treatment of alcoholic fatty liver is based on the Adip/CaMKK?/AMPK signaling pathway to improve the alcohol-induced steatosis.
