A Comparative Study On The Commercial Grade And Quality Of Peucedani Radix Medicinal Materials And Pieces

Objective:1.Based on the methods of TLC,HPLC and fingerprint,a systematic evaluation system for the quality comparison of Qianhu medicinal materials was established,and the methods suitable for this study were explored to provide the basis for the quality comparison of different specifications and grades of medicinal materials and pieces.2.Carry out systematic quality test on the graded samples of Qianhu medicinal materials and decoction pieces,and compare whether there are differences in the quality of samples from different places of origin and different specifications,so as to provide reference for the rationality of the classification of Qianhu medicinal materials and decoction pieces.Method:The TLC method was used to qualitatively identify the medicinal materials with the reference materials of Praeruptorin A,Praeruptorin B and Praeruptorin E.The water,ash and alcohol soluble extracts were detected according to the requirements of Pharmacopoeia 2015.The three coumarin components and fingerprints of peucedane and its pieces were determined by HPLC Analysis and comparison of the data obtained from grade II herbs.Result:1.The classification of the medicinal materials of Qianhu is relatively consistent,which is divided into unified,large,medium and small;the pieces of decoction are mainly divided into selected and unified,the classification method is relatively simple,screening and classification.A total of 44 batches of medicinal materials of different specifications and grades were collected,and 132 batches of pieces were screened and graded by ourselves.2.According to the overall analysis data,among the different specifications and grades of Qianhu medicinal materials,the average value of total ash is the highest in datiao and the lowest in Tonghuo.Compared with Tiaohu,the total ash content ofTiaohu is greater than that of Tiaohu.However,the average value of acid insoluble ash is higher than that of Tiaohu.The results showed that there was no certain regularity among different grades of herbs,but from the overall average value,the content of lixivium was slightly higher than the control.In the determination of three coumarin components,the content of B in commodity grade of Qianhu medicinal materials showed a certain regularity with the place of origin.In Anhui Province,the content of B in prenatal Hu medicinal materials showed a negative correlation with the commodity grade(except for the control of goods),that is,the content of B in large < medium <small;in Zhejiang Province,the content of B in Qianhu medicinal materials showed a positive correlation with the commodity grade,that is,large > medium > small > control of goods.Regardless of the origin,according to the overall content data,the mean value of the total content of three coumarins among the commodity grades of Qianhu medicinal materials is large(1.35%)< medium(1.36%)< small(1.40%)< unified(1.46%).The similarity of fingerprints of all producing areas is greater than 0.9,and 6common peaks are identified in total.Through comparison with the mixed control,3 of them are identified.Peak 2 was Praeruptorin A,peak 4 was Praeruptorin B,and peak 5was Praeruptorin E.3.In the comparison of the quality of different specifications and grades,it is found that,on the whole,the average value and grade of the extract of the slices show regularity: first grade > second grade > third grade,and the average value of the whole product is close to the second grade.There is no obvious regularity between the total ash content of the decoction pieces,but there is a certain regularity between the acid insoluble ash content and the grade,that is,the first grade < the second grade < the third grade.The control of goods is between the second level and the third level,which is closer to the second level.As a whole,the three coumarin contents in the commercial grades of Qianhu decoction pieces were all grade I < grade II < grade III.The similarity of fingerprints of all samples was greater than 0.9.4.After comparing the grade quality of medicinal materials and pieces,it is foundthat there is a certain correlation between the change rule of grade and characters.Due to the influence of human factors,there is a certain degree of cross in the diameter of the root head of the three grades,and the quality difference is not obvious.But there are some differences between Tiaohu and Tonghuo.After screening and grading,it was found that the contents of extractives and coumarins changed regularly.Conclusion:1.There is a certain scientific rationality in the classification of specifications and grades of existing medicinal materials on the market.It is suggested to standardize the appearance index of each grade,which can provide reference for the classification of commodity grade.2.It has certain rationality and regularity to divide the specification grade by sieving.The qualified samples can be further collected for research to clarify the internal division index.
