Objective: To investigate the expression level of miR-29 b in CD19+B lymphocytes of patients with chronic lymphocytic leukemia(CLL)and its correlation with the prognostic indicators that have been identified so far,and to explore the diagnostic and prognostic value of miR-29 b in CLL.,.Methods:15 ml peripheral blood samples from healthy control(n = 40)and CLL(n = 80)patients were collected,and CD19 + B lymphocyte RNA was extracted after sorting with CD19 magnetic beads.The expression of miR-29 b was detected by TapMan probe CLL patients were grouped according to factors such as staging(Binet and Rai staging),IGVH mutation,genetic prognosis grouping,CD38,ZAP70,P53 abnormality(mutation or deletion),ATM deletion,IPI score and other factors,and miR-29 b was detected Whether there is a difference in expression,analyze the expression difference of miR-29 b in different groups,and use Cox regression to perform multivariate survival analysis on the above prognostic indicators and miR-29 b.Results:(1)In 80 CLL patients and 40 healthy people,the expression of mir-29 b increased in CLL patients(P < 0.001);(2)The expression level of miR-29 b in IGVH mutation group was lower than that in non mutation group(P < 0.05),ATM deletion group was lower than that in non deletion group(P < 0.05),p53 abnormality group was lower than that in normal group(P < 0.05),IPI high-risk group was higher than that in low-risk group(P < 0.05),but according to Binet and Rai stages,CD38,ZAP70 There was no significant difference in the expression level of mir-29 b in different groups of cytogenetics(P > 0.05).(3)The results of survival analysis showedthat TFs in the low expression group was significantly lower than that in the high expression group(P< 0.05).The Cox multivariate analysis of mir-29 b combined with IGVH,ATM,p53,IPI and other prognostic factors showed that the high risk group of IPI and the low expression of mir-29 b were the independent adverse prognostic factors affecting the survival of CLL patients(all P < 0.05).Conclusion:The expression level of miR-29 b in CLL patients is significantly higher than that in the normal control group.The low expression of miR-29 b is an independent poor prognostic factor for CLL.
