Objective:To prepare a compound milk-derived probiotic camel milk(containing 10kinds of lactic acid bacteria and 4 kinds of yeast)in the laboratory,and first observe its effect on the intestinal mucosal barrier function of diabetic rats,and secondly,14 kinds of fermented milk-borne probiotics were used to observe on colonization ability,antibiotic resistances safety andα-glucosidase inhibition rate.Methods:1.Resuscitate lyophilized powder of probiotics,determine viability and count of viable bacteria.Preparation of compound probiotic camel milk.Fifty SPF Wistar rats were given 45%high-fat diet and injected with streptozotocin to establish a type 2 diabetic rat model.Wistar rats of the same week age were used as normal control group,model group,metformin group(0.3g/kg),compound probiotic low-dose group(1×10~8 lactic acid bacteria+1×10~6 yeast)CFU and high-dose group(1×10~100 lactic acid bacteria+×10~8 yeast)CFU,a total of five groups,intervention for 6 weeks.Measurement of relevant blood glucose and lipid indicators:body weight(BW),fasting blood glucose(FBG),glucose tolerance(OGTT),glycated hemoglobin(HbAlc),C peptide(C-peptide),total cholesterol(TC),triglycerides(TG),Low density lipoprotein(LDL-C)and high density lipoprotein(HDL-C).Indicators affecting the function of the intestinal barrier:diamine oxidase,D-lactic acid,bacterial endotoxin,TNF-α,IL-1β,IL-6,pancreatic tissue,colon tissue,intestinal tight junction protein and mucin expression.2.To investigate the probiotic characteristics and safety of surface lactic acid bacteria and 4 types of yeasts,such as surface hydrophobicity,self-aggregation,and adhesion,and to compare the inhibition rates ofα-glucosidase by 14types of fermentation bacteria.Results:1.After 6 weeks of milk-derived complex probiotics intervention,FBG(P<0.01),OGTT(P<0.01),and HbAlc(P<0.05)in diabetic rats were significantly reduced,and C-peptide content was increased(P<0.05);HE staining of pancreatic tissue showed that the number of islets in diabetic rats in the high-dose group increased,the volume increased,and the number of islet cells increased.2.Compound probiotics can significantly reduce the contents of TG(P<0.001),TC(P<0.01),and LDL-C(P<0.01)in diabetic rats.3.Compound probiotics reduce the levels of DAO,D-lactic acid,bacterial endotoxin,TNF-α,IL-1β,IL-6 in serum,and relieve endotoxemia and chronic low-grade inflammation.4.Compound probiotics can significantly increase the mRNA and protein expressions of tight junction protein and mucin in the intestine of diabetic rats.5.10 kinds of lactic acid bacteria and 4 kinds of yeasts have good surface hydrophobicity,self-aggregation performance,adhesion and safety.The bacteria with a higherα-glucosidase inhibition rate are:L.kefianofaciens、L.lactis、I.orientalis.Conclusion:Xinjiang probiotic milk-derived probiotics may regulate mRNA of intestinal tight junction protein in diabetic rats.And protein expression,reduce intestinal permeability,improve endotoxemia and alleviate chronic low inflammation,lower blood lipids and lower blood sugar to exert its anti-diabetic effect.
