Objective: To explore the distribution characteristics of MICA gene in Hui,Kazakh and Uyghur populations in Xinjiang population;the association between MICA gene polymorphism and cervical cancer;the expression and localization of MICA protein in cervical cancer tissues and cervical cells.Methods:(1)Collecting DNA from blood samples,using PCR-SBT technology and PCR-SSP technology to detect the expression of MICA gene and MICA-STR in Xinjiang population(83 cases of Xinjiang Hui,120 cases of Kazakhs,183 cases of Uygur)and case control population(137 cases of cervical cancer patients and 120 healthy controls matching demographic data),and(2)The expression of MICA protein in cervical squamous cancer(CSCC),squamous intraepithelial lesions(SIL),and cervicitis issues was detected by immunohistochemical method.(3)The expression and positioning of MICA in Cervical cancer C33 a,Siha and Hela cell lines,and H8 of the normal cervical epithelial cell line were detected by means of combination of immunofluorescent chemistry and laser confocal microscopy.Results:(1)A total of 21 MICA alleles were detected in the Xinjiang population,5 MICA-STR different types were detected.Two new alleles were found among the Kazakh population in Xinjiang,which has been officially named MICA*088N and MICA*029:02 by the World Health Organization's Naming Committee.(2)The frequency of MICA* 008: 01 and MICA-A5.1 was higher in women than men,and the difference was statistically significant(P<0.05).(3)MICA*008:01 is the most common allele of Hui,Kazakh and Uyghur in Xinjiang,the distribution frequency is different,the frequency of MICA*016 alleles in the Kazakh group is significantly higher than that of the Uygurs,the frequency of MICA*019 alleles in the Hui population is significantly higher than that of the Uygurs,and the frequency of MICA:010 alleles in the Hui population is significantly higher than that of the Kazakh,with a statistically significant difference(P<0.05).MICA-STR type,the most common type of Hui is A5,Kazakh and Uyghur most common type A6,Hui population A5 frequency is higher than the Kazakh population,the difference is statistically significant(P<0.05).(4)The distribution of MICA alleles in the population of Xinjiang was compared with that of other populations,The frequency of MICA*009:02 alleles in the Kazakh were higher than those of Hunan Tujia,Northern Chinese,Korean,Thai,South American Indian,Japanese and British.The frequency of MICA-010 alleles in the Kazakh and Uyghur population was lower than that of Han Chinese,Koreans,Thais and South American Indians in northern China.MICA-STR compared with Hunan Tu,Northern Chinese,A4 type frequency in Xinjiang Hui population is lower than Hunan Tujia,Xinjiang Kazakh population A5 type frequency is lower than Hunan Tujia family and Northern Chinese,the difference is statistically significant(P<0.05).(5)The frequency of A4 in the cervical cancer group was significantly lower than that in the healthy control group,and the differences were statistically significant(P = 0.0369,OR = 0.498,95% CI = 0.256~0.969).(6)The main expression of MICA protein is located in the cytoplasm,and the positive expression rate in cancer tissues and SIL tissues is significantly higher than that in cervicitis issues,the difference is statistically significant(P<0.05);there is no statistical significance between other clinical pathology parameters.Conclusions:(1)MICA gene has polymorphism characteristics in the Xinjiang population(Hui,Kazakh,Uyghur).MICA*008:01 is the most common allele in the above-mentioned population,the distribution frequency is different.MICA-STR,the most common type of Hui is A5,the most common type of Kazakh and Uyghur type A6,Hui population A5 frequency is higher than the Kazakh population.MICA*008:01and A5.1are more frequent among women than men in Xinjiang.(2)MICA alleles have unique characteristics in the population distribution in Xinjiang region,compared with other groups,the distribution of MICA alleles in Xinjiang and Hunan Tujia,Han Chinese,Koreans,Thais,South American Indians,Japanese,British,MICA 009:02,MICA:010 and other alleles in Xinjiang region population frequency difference from others.MICA-STR type compared with Hunan Tujia family,Han in northern China,Xinjiang Hui population A4 type frequency is lower than Hunan Tujia family,Xinjiang Kazakh population A5 type frequency is lower than Hunan Tujia family and han in northern China.(3)The frequency of A4 in the cervical cancer group was significantly lower than that in the healthy control group,suggesting that A4 allele may reduce the incidence of cervical cancer.(4)MICA protein with the exacerbation of cervical tissue disease gradually increased,indicating that MICA protein may be involved in the occurrence of cervical lesions.
