Objective:To analyze the expression of TIGIT on T cells in peripheral blood and cancer tissues of cervical cancer patients with cervical squamous cell carcinoma and to explore the correlation between TIGIT and clinical characteristics of cervicalsquamous cell carcinoma patients,so as to provide a basis for the development ofcervical cancer immunotherapy.Methods:52 patients with cervical cancer first diagnosed in the Affliated Tumor Hospital of Xinjiang Medical Universityfrom October 2017 to June 2019,and 18 healthy volunteers were collected as controls.The expression of TIGIT on T cells in peripheral blood and cancer tissues was detected by flow cytometry.The expression of TIGIT on T cells in peripheral blood,peripheral blood and cancer tissues of patients with cervical cancer was compared with that of the control group.The relationship between the expression of TIGIT and clinical features of patients with cervical cancer was analyzed..Results:1 The expression levels of TIGIT on CD4+T cells and CD8+T cells in patients with cervical cancer and healthy controls were(24.10±8.02)%,(50.69±18.32)%,(18.21±4.73)%and(36.00±10.38)%,respectively.The expression of TIGIT on CD4+T cells and CD8+T cells in patients with cervical cancer were higher than those in healthy controls(t=-3.740,p=0.000;t=-4.163,p=0.000).The difference between the two groups was statistically significant.2 The TIGIT expression level of CD4+T cellsin peripheral blood of patients with cervical squamous cell carcinoma was(24.10±8.02)%,and that of cancer tissues was(57.90±16.77)%,and the difference was statistically significant(P=0.000).The TIGIT expression level of CD8+T cells in peripheral blood of cervical cancer patients was(50.69±18.32)%,and that ofcancer tissues was(73.72±23.44)%,the difference was statistically significant(P=0.000).3The expression level of TIGIT on CD4+T cells in cancer tissues was relatedto age and menopausal history,The expression level of TIGIT on CD4+T cells inpatients with age?52 years old was lower than that in patients with age<52 years old,and thedifference was statistically significant(P=0.005).The expression level of TIGIT in CD4+T cells of postmenopausal group was lower than that of non-menopausal group,and the difference was statistically significant(P=0.025).Theexpression of TIGIT on CD8+T in cancer tissues was significantly different among the gross types oftumors(P=0.046).4 The expression level of TIGIT on CD8+T cells in peripheral blood was related to age,menopausal history,abortion history and HPV infection.The expression level of TIGIT in CD8+T cells of cervical cancer? 52 years old group was higher than that in < 52 years old group,and the difference was statistically significant(P=0.036).The expression level of TIGIT in CD8+T cells of postmenopausal group was significantly higher than that of non-menopausal group(P=0.014).The expression level of TIGIT in CD8+T cells of abortion history group was higher than that of non-abortion group,and the difference was statistically significant(P=0.027).The expression level of TIGIT in CD8+T cellsof HPV infectiongroup was significantly higher than that of non-HPV infection group(P=0.008).However,there was no significant difference in the expression of TIGIT in peripheralblood andcancer tissues in tumor size,differentiation,clinical stage and other factors.Conclusion:The expression of TIGIT in peripheral blood of patients with cervical cancer is higher than that of healthy controls,but lower than that of cancer tissues.The expression of TIGIT in peripheral blood is affected by age,menstrual status,HPV infection and abortion history,which can provide reference basis for immunotherapy of cervical cancer.
