Objective: To explore the repair of cartilage defect with ECM-PCL-HA scaffold co-cultured synovial mesenchymal stem cells(SMSCs)and chondrocytes.Methods: The adipose mesenchymal stem cells transfected with TGF-? 3 by lentivirus were cultured into PCL-HA scaffolds,and then the cells were removed after the formation of extracellular matrix.The SMSCs and chondrocytes were co-cultured in ECM-PCL-HA scaffold.Then cell-scaffold composite was planted into the knee joint defects of New Zealand white rabbits.At 3 and 6 months,the materials were taken for macroscopic score,histological score,Hematoxylin Eosin staining,Safranine O-fixation green staining and type II collagen immunohistochemical staining respectively to observe the repair effect.Results: In the third month,the SMSCs and chondrocytes co-cultured in ECM-PCL-HA group can achieve good repair effect,which is significantly better than other groups.HE staining showed that PCL-HA scaffolds had cell adhesion and growth,blood vessels grew in the scaffolds,and PCL-HA scaffolds were well combined with surrounding normal bone and cartilage.Immunohistochemical staining showed the expression of type II collagen in new cartilage and scaffold.Conclusion: PCL-HA scaffold has good biocompatibility in vivo and can be used as a good cartilage tissue engineering material;extracellular matrix can promote cell adhesion and adhesion,promote the combination of cell material complex with surrounding bone and cartilage,and shorten the healing time of cartilage defect,which has certain clinical guiding significance.
