Objective: To inves tigate the changes and clinical signi fi cance of Tim-3/Galec tin-9 and Th1,Th2 rel ated cyt okines interferon gamma(IF N-?)and int erleuki n 4(IL-4)in CLL.Methods : 35 pat ients dia gnos ed with CLL and 30 hea lthy pers ons were select ed as the control group.There were 26 males and 9fem ales in the CLL group.The ave ra ge age of males was(62.19±13.64)yea rs and the average age of fem ales was(63.44±7.55)years.The re was no stati stica lly significant difference in age and gender betw een groups(P>0.05).Flow cyt om etry was used to det ect the expres sion levels of Th1 and Th2 and Tim-3 on Th1 and Th2.The changes of IL-4,IF N-? and Galecti n-9 were det ected by enz ym e-l inke d immunos orbent ass ay(ELIS A).Res ults :(1)Th1,Th2,Th1/Th2,Th2+ Tim-3 in the CLL group had no statis tical significance with the control group(P>0.05).Th1+Tim-3 and Th1/Th2+ Tim-3 in the CLL group were higher than thos e in the control group(P<0.05).(2)Gal ecti n-9 i n the CLL group was higher than tha t in the control group(P<0.05).IFN-? and IF N-?/IL-4 in CLL group were lowe r tha n thos e in control group(P<0.05).(3)According to Rai stage CLL group,Galec tin-9,Th1+ Tim-3 expres sion gradually increas ed with the higher stage(al l Pvalues <0.05).The expres sion of IF N-?/IL-4(%)dec re ased gradually with the higher stage(all Pvalues <0.05).(4)Th1+Tim-3 was negatively correlated with IN F-?(P=0.001,r=-0.323).Conclusi on: The combinati on of Tim-3 and Galectin-9 in Th1 cells with high expres sion in CLL patie nt s affects Th1 function.As the stage is higher,the secreti on of IF N-? is significantly reduced,and theimbalance of Th1/Th2 leads to changes in CLL microenvironment,which affects tumor cells.Cle arance.The Tim-3/ Gale ctin-9 pat hw ay may further affect the function of Th1 cells,the reby further regulat ing the leve l of IF N-?,leading to immune escape of tum or cells.
