Expression And Correlation Of 5-HT,DA,IL-6,TNF-?in Cia With Depression Rats

Objective: To investigate the expression and correlations of 5-HT,DA,IL-6,and TNF-? in the brain tissue and serum of collagen-inducedarthritis(CIA)rats with depression model,and to preliminary explore r heumatoid arthritis The pathogenesis of depression.Methods: Eighty wis tar rats were randomly divided into four groups after normal feeding fo r one week,with 20 rats in each group: blank group,CIA group,depre ssion group,and model group.The CIA rat model was combined with c hronic unpredictable mild stimulation(CUMS)to establish a CIA with d epression model.After modeling,sugar water consumption and tail-hangi ng experiments were performed to determine the behavior of rats,obser ve the synovial pathology of rats,and measure 5-HT,DA and IL-6 in b rain tissue and serum by enzyme-linked immunosorbent assay(ELISA).TNF-? content.Results:(1)Comparison of 5-HT and DA levels: Compar ed with the content in the brain tissue,compared with the blank group,the 5-HT and DA contents in the brain tissue of the model group,dep ression group and CIA group were reduced,and the 5-HT content in th e model group was lower than that in the depression group.And the CI A group decreased,the difference was significant(P<0.05);contrust wi th the DA,5-HT content in serum,compared with the blank group,mo del group and CIA group,the 5-HT and DA content in the depression g roup decreased,and The model group was lower than the CIA group,a nd the difference was statistically significant(P<0.05);(2)Comparison ofIL-6 and TNF-? levels: brain tissue content comparison,contrust withthe blank group,the model group,depression group and CIA group brai n tissue The levels of IL-6 and TNF-? increased,and IL-6 in the mode l group was higher than that in the CIA group.TNF-? in the model gr oup was higher than that in the depression group and the CIA group,a nd the differences were statistically significant(P<0.05);Compared withthe blank group,the serum levels of IL-6 and TNF-? in the model gro up,depression group and CIA group were increased compared with the blank group,and the IL-6 and TNF-? levels in the model group were h igher than those in the CIA group.All were statistically significant(P<0.05).(3)Correlation analysis: Blood IL-6 has a positive correlation withblood DA,blood TNF-? has a negative correlation with brain 5-HT,an d brain IL-6 has a negative correlation with brain 5-HT.Conclusion: In creased cytokine levels and decreased monoamine neurotransmitters in br ain tissues may be involved in the development of CIA with depression.
