Oxymatrine(OM,molecular weight 282)which extracted from Sophora flavescens has an antiviral and anti fibrosis effect,can significantly improve the patients after radiotherapy and chemotherapy leukopenia effect.Oxymatrine has significant therapeutic effects on killing tumor cells and inhibiting tumor cell invasion,and is also widely used in clinical treatment of auxiliary anticancer drugs.The aim of the trial is to explored the anti-tumor mechanism of OM.ObjectiveTo investigate the effects of Oxymatrine(OM)on the expression of P21 in human gastric cancer cell line SGC-7901 and the mechanism of inhibiting the proliferation of human gastric cancer cells.Methods1.Gastric cancer cell line SGC-7901 was cultured in vitro.2.Flow cytometry was used to observe the inhibitory effects of Oxymatrine(2mg/ml、3mg/ml、4mg/ml)on the proliferation of SGC-7901 gastric cancer cells at different time of 24 h and 36 h.3.The expression of P21 gene in gastric cancer cell line SGC-7901 was observed by Western blot methods.ResultsCompared with the control group,different concentration(2mg/ml、3mg/ml、4mg/ml)OM to cell cycle arrest in G1/G0 phase(24h: 67.5%±0.1%,69.5%±1.4%,71.0%±1.0% Vs 58.6%±0.4%,P<0.05;36h: 68.5%±0.3%,71.9%±0.9%,78.0%±0.4% Vs 58.8%±0.1%,P< 0.05)the expression level of up regulation of cell cycle negative regulatory factor P21(P<0.05).ConclusionsOM may play a novel role in anti-tumor effect by blocking the cell DNA synthesis,and can up regulate the expression of P21 in G1/G0 phase of in gastric cancer cells.
