The Relation Between The Quantity Of ??T Cells Or Th17 Cells In Peripheral Blood Form Chb Patients And Clinical Condition

ObjectiveThe gamma delta T cells and Th17 cells in the role of CHB is unclear.The study is to analyze the frequency and cytokines secretion of peripheral blood gamma delta T cells and Th17 cells in HBV infection patients with different phases.Evaluatedthe correlation with liver function injury and related indicators,and to further explore the role of clinical and prognosis in CHB.MethodsIn this study,a total of 60 people,including 20 patients with moderately severe CHB,20 patients with mild CHB,20 patients with chronic asymptomatic hepatitis B carriers,was tested.HBV diagnosis and staging of standards accordance with the plan.20 healthy people(medical no abnormal findings,nearly 2 months without infection related history)was as control.Flow cytometry was used to detect the cells expression quantity of ??T cells,Th17 cells in peripheral blood and ELISA was used to detece the cells expression quantity of IL-17 cytokine in serum form 20 asymptomatic HBV carriers(ASC),20 CHB patients in low grade,20 CHB patients in moderate or severe grade and 20 health biood donors.The transaminase and bilirubin were detected by automatic biochemical analyzer,and HBV-DNA was detected by PCR-fluorescent probe.All the date was analyzed by the SPSS16.0,One-Way ANOVA analysis was used to analyze the difference in the cells expression quatity or the relationship,the Mann-Whitney U and Spearman were used to analyze the relation between the cells or the clinical paramteters,and correlation analysis.Results1.??T cells can detect in CHB patients and healthy blood donors,and with the increasing of the CHB level,??T cells expressing quantity increases gradually,the relation between CHB in moderate or severe grade>CHB in low grade>ASC>health biood donors,and there was a significantly statistic difference between the groups(P=0.024<0.05).2.There was significantly positive correlation between ??T cells and clinical indicators of TB or DB(P<0.05),and positive correlation between ??T cells and clinical indicators of ALT or AST,but the HBV-DNA load had no effect on the ??T cells expression quantity.3.Th17 cells can be detected in CHB patients and healthy blood donors,and with the increasing of the CHB level,Th17 cells expressing quantity increases gradually,the relation between CHB in moderate or severe grade>CHB in low grade>ASC> health biood donors,and there was a significantly statistic difference between the groups(P=0.018< 0.05).4.IL-17 cytokine can be detected in CHB patients and healthy blood donors,and with the increasing of the CHB level,IL-17 cells expressing quantity increases gradually,the relation between CHB in moderate or severe grade>CHB in low grade>ASC>health biood donors,and there was a significantly statistic difference between the groups(P=0.023 <0.05).5.There was significantly positive correlation between ??T cells and clinical indicators of ALT or AST and the HBV-DNA load(P<0.05),but no correlation between ??T cells and other immune cells.Conclusions1.Our studies confirmed that the frequency of gamma delta T cells and Th17 cells of patients with moderately severe CHB in peripheral amount is significantly higher than patients with mild CHB and healthy controls,the frequency of Th17 cells quantity and cytokines IL-17 levels by secreted is significant positive correlation with HBV-DNA loads.The results showed that gamma delta T cells and Th17 cells involved in the process of the development of CHB.2.The frequency of gamma delta T cells increases that is significant positive correlation with the serum ALT levels.The frequency of Th17 cells and the content of IL – 17 are significant positive correlation with serum ALT levels.The results showed that gamma delta T cells and Th17 cells participate in the process of hepatitis B virus(HBV)mediated liver pathological damage.3.With the increase of amount of gamma delta T cells express,the expression of CD4 + T is increase,and the correlation with HBV DNA load is not significant.The correlation gamma delta T cells with CD8 + T cells and non-specific NK cells are not significant.The results showed that the frequency of gamma delta T cells increases are correlation with assistive T cell immune response in patients with HBV infection.But the HBV-DNA load had no effect on the gamma delta T cellsproportion.4.The content of IL-17 cell cytokines of CHB patients in moderate or severe grade is significantly higher than CHB in low grade and ASC and health biood donors.This prompted that the Th17 may be involved in the process of the immune response caused by virus infection and tissue damage,and it is important in the pathogenesis of immune system.
