Part 1 The correlation between Glu R2 expression and pathological classification,onset age or total seizure frequency of intractable epilepsyObjectives: To investigate the expression levels of Glu R2 of different pathological types and analyse the correlation of the onset ages and total incidence frequency with the Glu R2 expression in primary intracable epilepsy patients.Methods: 62 cases of IEP admitted to the Department of Neurosurgery,General Hospital of Jinan Military Area during January 2008 to June 2015 were included in this work.These patients were diagnosed by brain MRI,PET-CT or VEEG and the lesions were located by ECo G and cut off.The cut samples were put into ice box and sent to Department of Pathology immediately for the next process.Previous researchers have identified these specimens according to the latest ILAE standard type.All we need to do is to use immunohistochemical techniques to deal with these samples and use optical microscope to observe and count the cells in low and high magnification.Then,the image processing software was used to collect and modify the image and statistics software were used to analyzed the datas.Results: scores of Glu R2 expression and the total incidence frequency had a significantly negative correlation,R=-0.479,P=0.008<0.05;Glu R2 expression scores and onset age have significant positive correlation,R= 0.561,P=0.002<0.05;There was no significant difference(P>0.05)of Glu R2 positive expression scores between every two pathological types of the patients in intractable epilepsy group.Each of intractable epilepsy group was significantly lower than that of the control group(P<0.05).Conclusions First,The expression of Glu R2 was negatively correlated with the total incidence,and was positively correlated with onset age.Second,The expression of Glu R2 in refractory epilepsy was significantly decreased compared with that in normal control group.Third,There was no significant difference in Glu R2 expression among different pathological types.Part2 Comparison of NMDAR1 expression in cerebral cortex of IEP with and without cognitive impairment.Objective: To observe the differences of NMDAR1 expression in the IEP with cognitive impairment,IEP without cognitive impairment and normal control group,and explore the significance of NMDAR1 expression changes for epilepsy with cognitive impairment.Methods: All the 62 patients with IEP and 5 cases of normal control group were evaluated by simple mental evaluation table(MMSE)and Memory and executive screening scale(MES),and screened 13 patients with cognitive impairment.All samples were treated with immunohistochemical technique and NR1 was labeled by antibody.The results were observed and the data were analyzed.Results: There were significant difference among groups of IPE without cognitive dysfunction,IPE with cognitive dysfunction and normal control(P<0.05).The order of the average score of NR1 expression was that IPE without cognitive dysfunction>IPE with cognitive dysfunction> normal control.Conclusions: Recurrent seizures caused a reduction in the expression of NMDA receptor NR1 subunit,and resulted in cognitive impairment.
