Objective To investigate the expression PI3K/AKT signaling pathway of its related factors(PI3Kp110α、p-mTORser2448、p-AKTser473、CyclinD1、CDK4、NF-кB、RAS、ERK1/2)in colon cancer and their association with other clinical pathological parameters,and analyzing of interaction of tow signaling pathways in colon cancer.Methods Selection surgical removal of the 65 cases of patients with colon cancer(colon cancer group)during in January 2013 to December 2014 of jinan central hospital affiliated shan dong university,the male 34 cases and the female 31 cases,during 32-88 years,the median ageof 66.There are 36 cases moderately differentiated adenocarcinoma and 29 cases poorly differentiated adenocarcinoma.Using the 2010 TNM staging standards promulgated by Union for International Cancer Control:stage I 3cases,stage II 25 cases,stage III 24 cases,stage IV 13 cases.There are 33 cases lymph node metastasis and 32 cases with no lymph node metastasis.All specimens of colon cancer group were confirmed by postoperative pathology for adenocarcinoma,and with complete clinical data,preoperative without any treatment.Another selection 15 cases colonic adenoma(colonic adenoma group): 8cases male,7cases female,during 34-82 years old,the median age of 62.The expression of PI3Kp110α、p-AKTser473、pm TORser2448 and CyclinD1、CDK4、NF-кB、RAS、ERK1/2were detected with immunohistochemical method in 65 cases of colon cancer(colon cancer group)and 15 cases of colonic adenoma(colonic adenoma group).Applying Statistical package of social science13 statistics the date.The association between the expressions and clinical pathological parameters were analyzed by the Mann-Whitney U test of nonparametric statistics analysis inspection level for α=0.05,when P<0.05 the difference was statistically significant.Results 1、The positive expressions of PI3Kp110α、p-AKTser473、p-mTORser2448、Cycl inD1、CDK4、NF-кB、RAS and ERK1/2were significantly higher in colon cancer than in colonic adenoma group(all P<0.05).2、In colon cancer group,there were no differences in expressions of PI3Kp110α、p-AKTser473、p-mTORser2448 between different genders 、ages、different degrees of differentiation,lymph node metastasis and TNM staging(all P>0.05);3、The positive expressions of CyclinD1 and CDK4were significantly higher in poorly differentiated adenocarcinoma than in moderately differentiated adenocarcinoma colonic(all P<0.05),the positive expressions of CyclinD1 and CDK4 were significantly higher in III+IV staging than in I+II staging(all P<0.05),but no statistical differences between different genders,ages and lymph node metastasis(all P>0.05).4、The expression of NF-кB and ERK1/2 were significantly higher in lymph node metastasis than in no lymph node metastasis(all P<0.05),but no statistical differences between different genders,ages and vary ing degrees of differentiation(all P>0.05).5、The positive expressions of RAS were signifycantly higher in poorly differentiated adenocarcinoma than in moderately differentiated adenocarcinomacolonic(all P<0.05),but no statistical differences between different genders,ages and lymph node metastasis and TNM staging(all P>0.05).Conclusion The abnormal expression PI3K/AKT signaling pathway of its related factors(PI3K p110α、p-AKTser473、p-mTORser2448、CyclinD1、CDK4、NF-кB、RAS、ERK1/2)are closely related with the occurrence and development of colon cancer,and the abnormal expression reveal these:1、The overexpression of PI3Kp110α、P-AKTser473、p-mTORser2448 are closely related with the occurrence and development of colon cancer.2、The higher positive expression rate of CyclinD1 and CDK4,the later stage of tumor and the worse differentiation in colon cancer.It’s reveal that the overexpression of CyclinD1 and CDK prompted further development and distant metastasis of colon cancer.3、NF-кB plays an important role in the development of colon cancer,especially in the lymph nodes metastasis and the TNM staging.4、The overexpression of RAS gene plays a very important role in the progress of the colon icadenoma and cancelations,and reveals poorly differentiated.5、The overexpression of ERK1/2 is closely related to the lymph node metastasis and TNMstages of colon cancer,it plays a promoting role in the occurrence and development of colon cancer.6、PI3Kp110αP-AKTser473、P-mTORser2448、CyclinD1、CDK4 、NF-кB、RAS andERK1/2 are significant in the diagnosis,treatment,prognosis assessment and new research targets of colon cancer.
