Objective:Organ preservation solution bacterial contamination is widespread in the field of organ transplantation,organ transplantation in patients with postoperative infection caused by its have a high fatality rate.In order to decrease the infection rate of the patients after renal transplantation,and explore the effective means of prevention,understand the antibacterial activity of cephalosporin which added to the kidney organ preservation solution advancely is high or low and whether there is damage to the kidney function,provide a theoretical basis for prevent infection from organ preservation solution contamination after renal transplantation effectively In the clinical,do this experiment.Methods:Formulate bacteria suspension with bacteria of logarithmic phase,put logarithmic phase suspension of staphylococcus epidermidis,staphylococcus aureus and e.coli in quadruplicate vaccination respectively in Tryptic Soy Broth(TSB)and University of Wisconsin solution(UW),and culture for 24 hours at the condition of 4?and 37?respectively,compare and analysis of growth situation of various bacteria,understand the effects of temperature and culture medium itself on the bacterial growth.staphylococcus epidermidis strains,Staphylococcus aureus strains and Escherichia coli strains were exposed to ceftriaxone and cefazolin at concentrations from 0 to 1000?g/mL under hypothermia and normothermic conditions in UW.To simulate programmed conditions(bacteria from organ preservation solution to the situation of the human body),add the epidermis staphylococcus vaccination in 0-1000?g/mL of cefazolin UW solution in train20 hours under 4?,then abandon to UW liquid reserves the strains with tryptone soy broth flushing strain 3 times to remove cefazolin and join with the volume of soy broth,tryptone in 37?44 hours,observe the bacterial growth.Through the cell viability experiment and caspase activity monitoring cefazolin endothelial cell toxicity to the pig kidneys.Results:Under the condition of without cephalosporin,under low temperature(4?),training for 24 hours,three kinds of bacteria,slow growth in soy broth tryptone bacteria increment is not obvious,but in the fluid of the UW,almost no growth,three kinds of bacteria bacterium quantity and vaccination before nearly equal;Under the normal temperature(37?),training for 24 hours,three kinds of bacteria to grow rapidly in soy broth tryptone,three kinds of bacteria in the fluid of the UW,total bacteria slow decline,18 hours before,after 18 hours total bacteria after a slight rise.Concentration of 1000?g/mL of cefazolin show bactericidal action of epidermis staphylococcus completely,even in cefazolin removal for a period of time,under the condition of epidermis staphylococcus will not enter the growth stage.Finally,the concentration of 1000?g/mL of cefazolin is no adverse effect to the pig kidney endothelial cells,the pig kidney endothelial cell viability not seen significant decline.Conclusions:Low temperature(4?)can reduce the sterilization efficiency of cefazolin,but to join the big dose of cefazolin UW solution,its total bacteria within 12 hours will drop to observe low limit(10~2 cfu/ml),did not increase the dose of cefazolin UW solution,the bacteria are not appear a large number of growth,but the bacteria will exist for a long time,once the bacteria will come into contact with the appropriate conditions recovery and a large number of proliferation,and join the large dose of cefazolin UW solution does not reduce its kidney preservation effect.Based on these findings,large dose of cefazolin can prevent application in low temperature organ preservation in order to reduce the organ preservation solution bacteria pollution,reduce the pollution of the organ preservation solution caused by the incidence of organ transplantation in patients with postoperative infection.
