PurposeAnalysis of the differential expression of the COX-2 gene and protein in familial clustering cerebral thrombosis,sporadic cerebral thrombosis and normal controls,and the association between rs20417,rs5275 and familial cluster cerebral thrombosis or sporadic cerebral thrombosis in the COX-2 gene,To provide a new theoretical research for the pathogenesis of family clustered cerebral thrombosis,and further explore the relationship between COX-2 and family clustering cerebral thrombosis.MethodTwenty-five cases of family clustered cerebral thrombosis,25 cases of sporadic cerebral thrombosis and 25 cases of normal control group were collected.The differences in COX-2 gene expression among the three groups were analyzed and real-time quantitative polymerase chain reaction(PCR)techniques were used to verify their differential expression.Differences in COX-2 gene expression among the three groups were compared using variance analysis and least significant difference method,whether statistically significant(P<0.05).The levels of COX-2 protein in the three groups were detected by ELISA.The differences between the three groups were statistically significant(P<0.05).The polymorphisms in the promoter region of COX-2 were examined to investigate the differences and associations of the genotypes of the rs20417 and rs5275 polymorphisms of the COX-2 gene in the three groups.Result1.There was no statistically significant difference in age,smoking history,drinking history,diabetes history between the family cluster cerebral thrombosis group,sporadic cerebral thrombosis group and the normal control group(P>0.05).2.COX-2 gene expression was significantly up-regulated in the family clustering cerebral thrombosis group compared with the normal control group,and the difference was statistically significant(P<0.05).3.There was no statistically significant difference in the protein expression of COX-2 in patients with family clustered cerebral thrombosis,patients with sporadic cerebral thrombosis and normal controls(P>0.05).4.There was no significant difference in the composition of COX-2 gene rs20417 and rs5275 genotypes in patients with family clustered cerebral thrombosis,sporadic cerebral thrombosis and normal population(P>0.05).Conclusion1.The COX-2 gene may be involved in the pathogenesis of cerebral thrombosis,and its up-regulation may increase the susceptibility of cerebral thrombosis.2.COX-2 protein expression level may not be related to the incidence of cerebral thrombosis3.The rs20417 and rs5275 genotypes of COX-2 gene may not be related to the incidence of cerebral thrombosis. |
