Objective: Based on the establishment of left atrial fibrillation model in chronic pacing dogs,investigate the expression and content of Notch-1 and L-type calcium channel ?1C subunits Cav1.2 and SERCA2 a in atrial myocytes of chronic atrial fibrillation,analyze the correlation between Notch-1 and calcium circulation channels.Methods: Healthy beagle dogs,weighing 18±2.66 kg,aged 7-9 years old,both male and female can be included.The experimental design and implementation were reviewed and approved by the Animal Ethics Committee.The experimental animals were randomly divided into 2 groups,the sham operation group(n=5): underwent minimally invasive thoracotomy but did not implant a pacemaker.Atrial fibrillation group(n=5):minimally invasive thoracotomy,left atrial appendage with pacemaker pacing 8week.Results: In the atrial fibrillation group,the atrial fibrillation can be successfully induced by rapid pacing of the left atrial appendage.After 8 weeks of experiment,the expression of Notch-1,Cav1.2 and SERCA2 a protein in the sham operation group was brown.Compared with the sham operation group,the expression of Notch-1 positive proteins in the atrial fibrillation group was dark brown,and the expression of Cav1.2 and SERCA2 a positive protein was light brown.The expression and concentration of Notch-1 positive proteins in the atrial fibrillation group were significantly increased,and the expression and concentration of Cav1.2 and SERCA2 a positive protein were significantly decreased,the difference was statistically significant(P<0.05).Pearson's analysis of the correlation between Notch-1 and Cav1.2 and SERCA2 a in the atrial fibrillation group showed that Notch-1 was negatively correlated with Cav1.2 and SERCA2a(r=-0.52,P=0.034;r=-0.66,P=0.025).Conclusion: Notch-1 is activated in atrial fibrillation and is associated withchanges in Cav1.2 and SERCA2 a,Notch-1 regulates calcium cycling pathways involved in the development of atrial fibrillation.
