Objective:To investigate the efficacy of Polymyxin B(PLO B)in the treatment of liver function damage caused by ?-Amanita peptide poisoning,and provide evidence for clinical application.Methods:20 New Zealand purebred white rabbits were divided into 4 groups,the first group was the blank control group;the second group was the poisoning model group,and the amanita toxin was used to administer the stomach;the third group was the poisoning model group plus the silibinin(In the group of Silibinin SIL,after intragastric administration of amanita toxin,the intraperitoneal injection was performed with silybin;the fourth group was treated with polymyxin B in the poisoning model group,and was administered with amanita toxin.Intravenous bolus injection of colistin B injection.Then the liver function and coagulation index were measured,and the statistical analysis was carried out according to the obtained data.The degree of damage of hepatocytes was observed by HE staining and immunohistochemistry,and the damage of hepatic function of ?-amanita peptide poisoning rabbit was investigated.Efficacy analysis.Results:(1)Alanine aminotransferase-infected New Zealand purebred white rabbits treated with polymyxin B and silybin,the activity of alanine aminotransferase and aspartate aminotransferase in rabbit serum was significantly reduced,total gallbladder The erythrocyte index was significantly lower,and the ALT,AST,and TBIL measurements in the polymyxin B treatment group were lower than those in the silybin treatment group,which was close to the normal value;(2)HE staiuing,polymyxin B and The degree of liver damage in the silybin treatment group was lighter than that in the untreated New Zealand purebred white rabbits,and the degree of liver damage in the polymyxin B treatment group was higher than that in the silybin treatment group.The degree of tissue damage was light;(3)immunohistochemical staining,the number of hepatocyte apoptosis in the polymyxin B and silybin treatment group was observed compared with that of untreated New Zealand purebred white rabbits.The number of hepatocyte apoptosis in the polymyxin B treatment group was less than that in the silybin treatment group.Conclusion:The above experimental results indicate that polymyxin B has the same protective effect on the liver of ?-Amanita peptide poisoning mice as the silybin,and the protective effect of polymyxin B is higher than that of silybin.
