Objectives:To explore the correlation between the expression level of MMRN1 gene with the clinical prognosis of patients with acute leukemia.Methods:1 The correlation between MMRN1 gene expression levels and clinical features in 31 AML patients and 19 ALL patients was analyzed.Using GUSB as an internal reference gene,the relative quantitative method 2-AACT was used to calculate the relative expression of the gene.The AML samples were divided into two groups according to the median(1.25)of MMRN1 expression,MMRN1 expression?1.25 for high expression MMRN1 group(17 cases),MMRN1 expression level<1.25 for low expression MMRN1 group(14 cases).Similarly,ALL samples were divided into two groups according to the median(3.87)of MMRN1 expression values.MMRN1 expression level>3.87 was highly expressed in MMRN1 group(10 cases),MMRN1 expression level<3.87 was low expression MMRN1 group(9 cases).Statistical analysis was performed using R version 3.4.1 software.The clinical characteristics of MMRN1 expression levels were compared between the two groups,and the difference was statistically significant.Univariate logistic regression was used to study the correlation between MMRN1 gene expression(as a categorical dependent variable)and clinical features.P<0.05 was considered to be statistically significant,and P>0.05 was considered to be statistically insignificant.2 The correlation between MMRN1 expression and clinical features and prognostic significance in 109 patients with AML were analyzed.According to the median(5.52)of MMRN1 expression,AML samples were divided into two groups.MMRN1 expression level>5.52 was highly expressed in MMRN1 group(54 cases),MMRN1 expression value<5.52 was low expression MMRN1 group(55 cases).Statistical analysis was performed using R version 3.4.1 software.The clinical characteristics of MMRN1 expression levels were compared between the two groups,and the difference was statistically significant.Univariate logistic regression was used to analyze the association between MMRN1 gene,immunohistochemical MPO,CG risk stratification and clinical features.Multivariate survival analysis was performed using the COX proportional hazard model,Kaplan-Meier method for OS analysis and survival curves,and the OS rate comparison between groups was performed by Log-rank test.P<0.05 was considered to be statistically significant,and P>0.05 was considered to be statistically insignificant.Results:1 In this study,the expression of MMRN1 was not correlated with clinical features[age,gender,WBC,HB,PLT,bone marrow blasts,risk stratification(AML)]in 31 patients with AML and 19 patients with ALL(P>0.05).2 109 patients with AML in the database2.1 Patients with high MMRN1 expression had higher MO counts than patients with low expression(P<0.0001).2.2 The expression of MMRN1 was significantly correlated with age(P=0.027)and MO(P<0.0001).There was no significant correlation between gender,peripheral blood WBC,HB,PLT,LY,and MMRN1 expression levels.2.3 Age(P=0.020)was significantly associated with CG risk stratification.Gender,peripheral blood WBC,HB,PLT,MO,LY,and MMRN1 continuous expression levels and MMRN1 expression levels were not significantly associated with CG risk stratification.2.4 MMRN1 expression levels(continuous)(P=0.023)and LY(P=0.002)were significantly correlated with MPO.Age,gender,peripheral blood WBC,HB,PLT,MO,and MMRN1 expression levels were not significantly associated with MPO.2.5 Survival analysis showed that the expression of MMRN1(P=0.038)and age(P<0.001)were significantly correlated with the overall survival of AML.Moderate(P=0.041)and poor(P=0.013)of CG risk stratification were significantly associated with overall patient survival.There was no significant correlation between gender,peripheral blood WBC,HB,PLT,MO,LY and MMRN1 expression levels and MPO and overall survival.After adjusting for relevant factors,there was no significant correlation between the continuous expression of MMRN1 and the overall survival of patients.The level of MMRN1 expression was significantly correlated with the overall survival of patients.2.6 The MMRN1 high expression group was significantly different from the low expression group(P=0.038),and the MMRN1 high expression group had lower overall survival rate than the low expression group.Conclusions:1 MMRN1 can be used for prognosis evaluation of AML patients,which is a factor indicating poor prognosis of AML.2 Age is a factor affecting the prognosis of patients with AML.MPO can not be used as a prognostic factor for AML.3 There was no significant relationship between WBC,HB,PLT,MO and LY in peripheral blood and AML patients.
