Objectives:To analysis the 16S rRNA of gut microbial in fecal and polyp tissues from CAP patients,then analyze metabolites in feces.To find the characteristic gut microbial and metabolites from the investigation of polyp tissues and feces of CAP patients,in order to further explore the correlation between gut microbial and metabolites.Methods:Collected fecal and tissues from 30 CAP patients(including 23 cases of tubular adenoma,6 cases of villus-tubular adenoma,and 1 case of villous adenoma)and 30 healthy volunteers.1.16S rRNA analysis:The genomic DNA was extracted from feces and tissues,then amplified V3-V4 region of 6S rRNA gene.After amplification,using high-throughput sequencing to obtain Clean Data,splicing sequences,species classification,and abundance analysis.2.Metabolites analysis:The collected feces were dissolved in dH2O,the content of short-chain fatty acids(SCFA)in the feces were determined by iron chromatography analyzer after filtered.The collected feces were dissolved in methanol,the content of bile acids and conjugated linoleic acids(CLA)in the feces were determined by UPLC-MS/MS analyzer after filtered.Results:1.16S rRNA analysis:A total of 5,994,516 high-quality sequences were obtained for all samples,and the coverage was over 99.9%.There was no difference between two groups in fecal group(CAPs group and HCs group).In the comparison of bacterial phylum,Cyanobacteria was differed between two group.There were 6 generas were significantly different between two groups,Erwinia,Weissella,Citrobacter,Lactobacillus,Peptoniphilus and Bacteroides.The index Chao and Ace of a diversity were statistically significant between the intestinal mucosa(CAPm group vs.HCm group),there were no significant differencebetween Shannon and Simpson index,and the difference between two groups were observed in ?diversity.The phylum difference between two groups was statistically significant between Bifidobacterium,Brevibacterium,Spirulina,Flexobacter,Thermi,Chlamydia,Saccharomyces cerevisiae and ODl.There were 68 generas were significantly different,the relative abundance of Bacteroides,Escherichia,Ruminococcus,Blautia,Dorea were increased in CAPm,and the relative abundance of Pseudomonas,Lactococcus,Geobacillus,Solibacillus were decreased.2.Metabolites analysis:Determination of SCFA in feces,in which acetic acid and butyric acid were higher in CAP group(P=0.003&P=0.000).The concentration of secondary bile acid(DCA,LCA)did not differ between two groups(P=0.22&P=0.45).The content of CLA's conjugate c9,t11-CLA was not different between two groups(P=0.234),while the content of t10,c12-CLA was significantly higher in HC group than in CAP group(P=0.013).Conclusions:1.The dominant genus in feces of CAP patients were Citrobacter,Bacteroides,2.The dominant genus in adenoma tissues of CAP patients were Pseudomonas,Lactococcus,Geobacillus,Solibacillus,Acinetobacter.3.In the feces of CAP patients,the content of acetic acid and butyric acid were increased,and the content of t10,c12-CLA were decreased,all these three metabolites were beneficial to intestinal health.4.There was no difference in secondary bile acid content in feces of CAP patients,which was harmful to intestinal health.
