Objective:One of the most important factors of cardiovascular disease in diabetes is energy metabolism dysfunction.Energy metabolism dysfunction causes the body homeostasis,hyperlipoidemia and hyperglycemia of the cardiovascular tissue damage its structure and function,and accompanied by cardiovascular systolic dysfunction.Thus,cardiovascular disease of diabetic occur metabolic disorders,while accompanied by systolic dysfunction.To investigate the effects of aerobic exercise and rosiglitazone on the expression of Peroxisome proliferator-acticated receptor ?(PPAR?)?Carnitine palmitoyltransferase-1(CPT-1)?Insulin-like growth factors-I(IGF-I)?Endothelin 1(ET-1)and Urotensin?(U?)in cardiac muscle and aorta of diabetic rats.To research themechanism of metabolic disorders and systolic dysfunction in diabetes.And explore the effects of aerobic exercise and rosiglitazone on metabolic and systolicfactor in cardiovascular of diabetic.Methods:1,Male Sprague-Dawley rats were divided into euglycemic control group and diabetic model group.The diabetic model group was intraperitoneally injected with streptozotocin(STZ)to induce diabetic model.The diabetic rats were randomly divided into three group: diabetic model group;diabetic model group with aerobic exercise;diabetic model group with aerobic exercise and rosiglitazone.The ras with exercise were forces to run on treadmill for 8 weeks.2,Measurement of blood glucose,glycated serum protein,triglycerides,total cholesterol and low-density lipoprotein of blood in rats.3,HE staining of cardiac and vascular.The tissue section was made by routine method for light microscope examination.4,The expression of metabolic shrinkage factors m RNA of cardiac and vascular was measured by RT-PCR.And the protein expression of metabolic shrinkage factors of cardiac and vascular was measured by Western Blot.5,A11 data were treated with SPSS 18.0.Measurement data were expressed as mean values--kstandard deviation.Results: 1,physiological status of rats:The body weight of the diabetic group was significantly lower(p ? 0.01).The body weight of the exercise group and the exercise dosing group increased.Heart weight of the diabetic groupreduced,heart weight ofthe exercise group and the exercise dosing group increased;Heart weight / body weight index of the diabetic group increased.The exercise group's reduced;OGTT in the diabetic group is abnormal.2,Changes in physiological and biochemical indexes:Blood glucose of diabetes groupwas significantly increased(p?0.01),Blood glucose ofthe exercise group and the exercise dosing group was significantly reduced(p?0.01),Glycated serum protein of diabetes groupwas significantly increased(p?0.01),Glycated serum protein of the exercise group and the exercise dosing groupwasreduced.Andtriglycerides,cholesterol and LDL levels of diabetic group are increased,and total cholesterol were significantly increased(p?0.01),Andcholesterol and LDL levels of the exercise group and the exercise dosing group was significantly reduced(p?0.01).triglycerides of the exercise dosing groupwasreduced.3,Cell morphology examination:Myocardial tissue of diabetic rats disorganized,and the cellstwisted and largen,the cell gap increased.The cell of the exercise group and the exercise dosing group still arranged neatly,cardiac hypertrophywas reduced,the cell gap becomes smaller.Endangium in the diabeticgroup thickened,and the endothelial cell swelling,the partof smooth muscle cellsderangement,endometrial cells of the exercise group and the exercise dosing group arranged in parallel,less damage.4,The changes of expressionabout metabolic factors: The expression of PPAR??CPT-1?IGF-1 in cardiac muscle and aorta of all diabetic group decreased compared with control group,the blood glucose and blood lipid increased.The expression of PPAR??IGF-1 in cardiac muscle and PPAR??IGF-1?CPT-1 in aorta were higher than that of diabetic group at the end of 8 weeks aerobic exercise,the blood glucose and blood lipid decreased.The expression of PPAR??IGF-I in cardiac muscle and PPAR??IGF-1?CPT-I in aorta were higher than that of diabetic group at the end of 8 weeks aerobic exercise and STZ injection,the blood glucose and blood lipid decreased.5,The changes of expressionabout contraction factors: The expression of ET-1?U? in cardiac muscle and aorta of diabetic group increased compared with control group.The expression of ET-1?U?in cardiac muscle and ET-1?U? in aorta were lowed than that of diabetic group at the end of 8 weeks aerobic exercise.The expression of ET-1?U? in aorta and U? in cardiac muscle were lowed than that of diabetic group at the end of 8 weeks aerobic exercise and STZ injection.Conclusions: 1,Metabolism in Diabetic Rats disorder,blood sugar and blood lipids elevated.Meanwhile,the metabolism and shrinkage factor of cardiac muscle and aorta changed,particularly cardiac and vascular metabolic factor PPAR?,CPT-1,IGF-I was significantly decreased,shrinkage factor ET-1 and U?was significantly higher.Will inevitably result in cardiovascular cell uptaking of glucose and lipids limited in diabetic rats,metabolism disorder,and excessive contraction of the blood vessels,myocardial blood supply shortage,and ultimately affect the cardiovascular systolic and diastolic function.2,Exercise can improve the glucose lipid metabolism and systolic dysfunction in cardiovascular of diabetic rats.Especially barriers of intaking sugars lipids in the cardiovascular cell and energy metabolism dysfuntion.While,exercise improve the abnormal vasoconstriction,increased myocardial blood supply.In addition,the effect on aorta is more obvious than on the myocardium.3,Effect of comprehensive intervention is more obvious in the cardiovascular metabolic disorders in diabetic rats,suggesting that comprehensive intervention is an effective way to improve cardiovascular metabolism.
