Glioblastoma is the most malignant brain tumor with poor prognosis.Its clinical course is usually rapid and fatal,with a median survival of less than 1 year.The current standard treatment for glioblastoma includes surgical resection within a safe and feasible range,followed by medication concomitant with radiotherapy.However,patients have a high probability of postoperative recurrence and a short recurrence time(usually within 8 months).The invasive ability of malignant glioma is an important factor in the tumor recurrence and distant migration.Tumor metastasis is the most common cause of death in cancer patients.In addition,since liver cancer has been ranked in the top five in China's cancer mortality rate,which poses a serious threat to the lives and health of our people.Therefore,in addition to the study of glioblastoma,the expression of PPFIBP1 protein in liver cancer,cirrhosis,hepatitis and normal liver tissues was further studied to explore the relationship between PPFIBP1 protein expression and liver cancer formation.The PPFIBP1 gene has been shown to be a cancer-related gene.The liprin-?1 protein encoded by the PPFIBP1 belongs to the leukocyte common antigen-associated(LAR)transmembrane tyrosine phosphatase interacting protein family.Studies have shown that the transfer-associated protein S100A4 interacts specifically with liprin-?1 in vivo,and this protein has been shown to induce invasiveness of primary tumors and promote tumor metastasis.However,there is currently no study of the PPFIBP1 in glioblastoma and liver cancer.We investigated the relationship between PPFIBP1 gene expression and migration or invasion of glioblastoma by scratch assays and transwell invasion experiments.Our results showed that the rate of migration and invasion in the PPFIBP1 overexpressed glioblastoma cells was much higher than that in the control group,and the results were reversed after PPFIBP1 silencing.After staining the liver tissue microarray by immunohistochemistry,we found that the expression of PPFIBP1 gene in primary hepatocellular carcinoma(HCC)was significantly higher than that of non-tumor liver tissue with signs of hepatitis or cirrhosis.It indicates that PPFIBP1 may be involved in the malignant transformation and development of hepatocellular carcinoma.The above results indicate that the PPFIBP1 can be used as a novel biomarker and therapeutic target for human glioblastoma and hepatocellular carcinoma.
