Armillaria mellea is a widely used medicinal fungus,mainly distributed in the northeastern region of China.In this paper,the identification of A.mellea,the examination of medicinal materials,the determination of heavy metals and beneficial elements,the fingerprints and the pharmacological effects of different extracts on rheumatoid arthritis were studied.The review section provides an overview of the chemical composition,pharmacological effects,and quality control studies of A.mellea.According to the quality control research overview,it can be found that A.mellea is unable to set standards.Since the Chinese patent medicines of the A.mellea group are all produced in the Jilin Province pharmaceutical factory,it is of great significance to establish the A.mellea standard in order to ensure the safety of the people's medicine and improve the quality of the Chinese patent medicine in Jilin Province.There is no report on the treatment of rheumatoid arthritis with A.mellea,so it is of great value to explore the different extracts of A.mellea for the treatment of rheumatoid arthritis.The research content is divided into four parts,namely,the evaluation of the authenticity of A.mellea,the examination of A.mellea,the evaluation of the effectiveness of A.mellea and the draft quality standards of A.mellea.In the authenticity evaluation of A.mellea,33 batches of 38 batches of A.mellea samples from different sources were identified for molecular identification,and four species of A.mellea were identified.All batches of A.mellea samples were also characterized for trait identification,microscopic identification and thin-layer identification,establishing identification methods and laying the foundation for A.mellea quality standards.In the study of A.mellea medicinal materials,the moisture,total ash,acid-insoluble ash,leachate,heavy metals,harmful elements and beneficial elements of different batches of A.mellea were tested.The moisture content of 38 batches of A.mellea samples was determined by toluene method,and the moisture content of A.mellea was not to exceed 14%.Determination of ash and acid-insoluble ash in 11 batches of A.mellea,the ash content of A.mellea shall not exceed 10%,and the acid-insoluble ash shall not exceed 0.43%.10 batches were randomly selected from the A.mellea for the determination of water-soluble extracts,and the water-soluble extract of A.mellea shall not be less than 25%.The contents of heavy metals and harmful elements such as lead,cadmium,arsenic,mercury and copper in different batches of A.mellea were determined.The results showed that the content was within the specified range.A.mellea was not enriched with heavy metals and harmful elements.Safety;The contents of beneficial elements such as calcium,magnesium,manganese,iron and zinc were also determined for different batches of A.mellea.The results showed that the content of magnesium in A.mellea was higher.In the evaluation of the effectiveness of A.mellea,the determination of the active ingredient content in A.mellea,the fingerprint and the therapeutic effect of A.mellea extract on rheumatoid arthritis were studied.The fingerprint similarity evaluation of 38 batches of A.mellea from different sources was carried out by adopting the Chinese Medicine Chromatographic Fingerprint Similarity Evaluation System 2004 A Edition.At the same time,the response surface analysis was combined with high performance liquid chromatography to optimize the optimal extraction process of adenine in A.mellea and determine its content.The fingerprint of A.mellea was established,8 common peaks were identified,and 1 peak was identified.The similarity of fingerprints of 38 batches of A.mellea samples was higher.Therefore,the fingerprint of A.mellea can effectively identify the authenticity and quality of A.mellea.It is of great significance to the establishment of regulatory and legal standards for A.mellea food and medicine.A.mellea was examined by measuring the degree of foot swelling in rats with adjuvant arthritis,systemic arthritis index score,body weight and body state changes,immune organ index,morphology of inflammatory ankle joint sections,and inflammatory factors in serum,to find out the effects of five different extracts on adjuvant arthritis rats and the mechanism of action.The results showed that the methanol extract of A.mellea had a significant inhibitory effect on paw swelling;The methanol extract of A.mellea can significantly inhibit the spleen and thymus atrophy in adjuvant arthritis rats,and inhibit the formation of rat sputum arthritis cells by inhibiting the secretion of serum inflammatory factors.Therefore,the methanol extract of the extract of A.mellea was most effective in the treatment of adjuvant arthritis rats.
