In recent years,with the accelerated pace of life and changes in the living environment,the incidence of kidney disease has increased year by year,and the types of simple kidney diseases and their complications have gradually increased.It has become a type of disease that threatens human health and cannot be ignored.The occurrence and development of kidney disease are affected by many reasons,and environmental factors are an important factor.How to test the impact of environmental factors more comprehensively on kidney disease has gradually become a hot spot in the study of kidney disease.In this paper,NRK-52 E was used as a model for rat renal tubular epithelial cells.NRK-52 E cells were cultured at different concentrations with glucose and connective tissue growth factor(CTGF)to detect their biological activities on NRK-52 E cells influences.The atomic force microscopy(AFM)was used as a detective technique to detect changes in cell morphology,height,Young's modulus and adhesion at the single cell level,which also combined and validated by biological analysis methods.The results of AFM test showed that after the stimulation of glucose(final concentration of 5.5,10,25 mM)and CTGF(final concentration of 0,2,10 ng / mL)for 72 h,the morphology of the cells gradually changed from elliptical to long fusiform.Moreover,the results shown that the heights of cells were increased,the adhesion of cells were decreased,the Young's modulus of cells were increased,and all showed a dose-dependent relationship.When the CTGF concentration reached 50 ng/mL and the glucose concentration reached 50 mM,the cell damage was severe and no longer changed.Western blotting test(Western Blot)showed decreased expression of E-cadherin,increased expression of ?-smooth muscle actin(?-SMA),fluorescent labeling shown rearrangement of cytoskeleton,and decreased cell viability by MTT assay.The above results indicated that CTGF or glucose stimulation can damage NRK-52 E cells and reduce the cell viability.In this paper,the combination of AFM mechanical detection technology and biological detection technology were used to identify the effects of different environmental factors on the biological activity of renal tubular cells,which mainly focused on cytoskeletal rearrangement and cadherin adhesion studies of cadherin-mediated cells.This research wish to provide new scientific ideas for the study of kidney disease.
