In the process of constructing the important functional tissue and organ,the research of the interaction of biomaterial-body is very critical,and has increasingly become an important frontier in tissue engineering,materials science and regenerative medicine.The control of cell fate through the regulation of physical factors has been a hot topic in the study of biomaterial-body(cell/tissue)interactions in recent years.Micro-patterning is a method that relies on physical topology to realize mechanical force to guide cell behavior.In recent years,micro-patterning technology has gradually matured and become a powerful means to study and control the fate of cells.By designing hydrophilic and hydrophobic surfaces of biomaterials and optimizing the micro-patternalized topological structure distribution of extracellular matrix proteins,the adhesion,growth distribution and orientation of stem cells can be effectively regulated,thus further influencing the behavior of directional differentiation of cells.By limiting stem cells to the micro-patternalized topological structure on the surface of biomaterials,the micro-patternalization of cells can be realized,so as to study the effects of micro-patternalization on the upstream cytoskeleton kinetics and downstream gene expression,and ultimately to realize the regulation of stem cells' stretching,migration,differentiation and other behaviors.In this study,an innovative micro-pattern system based on gelatine-calcium alginate was proposed.The micro-pattern system can realize 2D micro-patternization of different shapes and sizes,from single cells to multi-cells,from homogenous cells to heterogeneous cells,and thus realize the regulation of cell behavior.Objective and fluorescence staining results showed that the innovative micro-pattern system based on gelatin-calcium alginate can effectively restrict single cell to the square,rectangle,circle and triangle micro-pattern,multicellular can be effectively restricted to square,rectangle,circle,triangle,diamond,pentagram,grid,chessboard,stripes and grids micro-patterns,implements migration regulation of unicellular or multicellular and growth regulation of different kinds of multicellular.ALP and oil red O staining results show that the innovative gelatin-calcium alginatemicro-pattern's implementation of single bMSCs 2d micro-pattern has remarkable effect on the osteogenesis and adipogenic differentiation,the square and circular micro-pattern is more likely to promote bMSCs' s adipogenic differentiation,but rectangle and triangle micro-pattern is more easily to promote bMSCs' s osteogenesis differentiation.The innovative gelatine-calcium alginate micro-pattern system in this project is cheap,easy to obtain,easy to operate,non-cytotoxic and has excellent biocompatibility,which is helpful to study the effect of micro-pattern on stem cell limiting,migration and differentiation and the construction of artificial tissue of heterogeneous cells.
